2-methylacetoacetyl-coenzyme A thiolase (beta-ketothiolase) deficiency: one disease - two pathways

Background: 2-methylacetoacetyl-coenzyme A thiolase deficiency (MATD; deficiency of mitochondrial acetoacetyl-coenzyme A thiolase T2/ “beta-ketothiolase”) is an autosomal recessive disorder of ketone body utilization and isoleucine degradation due to mutations in ACAT1.
Methods: We performed a systematic literature search for all available clinical descriptions of patients with MATD. 244 patients were identified and included in this analysis. Clinical course and biochemical data are presented and discussed.
Results: For 89.6 % of patients at least one acute metabolic decompensation was reported. Age at first symptoms ranged from 2 days to 8 years (median 12 months). More than 82% of patients presented in the first two years of life, while manifestation in the neonatal period was the exception (3.4%). 77.0% (157 of 204 patients) of patients showed normal psychomotor development without neurologic abnormalities.
Conclusion: This comprehensive data analysis provides a systematic overview on all cases with MATD identified in the literature. It demonstrates that MATD is a rather benign disorder with often favourable outcome, when compared with many other organic acidurias.
Background 3-hydroxy-3-methylglutaryl-coenzyme A lyase deficiency (HMGCLD) is an autosomal recessive disorder of ketogenesis and leucine degradation due to mutations in HMGCL .
Method We performed a systematic literature search to identify all published cases. 211 patients of whom relevant clinical data were available were included in this analysis. Clinical course, biochemical findings and mutation data are highlighted and discussed. An overview on all published HMGCL variants is provided.
Results More than 95% of patients presented with acute metabolic decompensation. Most patients manifested within the first year of life, 42.4% already neonatally. Very few individuals remained asymptomatic. The neurologic long-term outcome was favorable with 62.6% of patients showing normal development.
Conclusion This comprehensive data analysis provides a systematic overview on all published cases with HMGCLD including a list of all known HMGCL mutations.
Timely recognition of threats can be significantly supported by security assistance systems that work continuously in time and call the security personnel in case of anomalous events in the surveillance area. We describe the concept and the realization of an indoor security assistance system for real-time decision support. The system consists of a computer vision module and a person classification module. The computer vision module provides a video event analysis of the entrance region in front of the demonstrator. After entering the control corridor, the persons are tracked, classified, and potential threats are localized inside the demonstrator. Data for the person classification are provided by chemical sensors detecting hazardous materials. Due to their limited spatio-temporal resolution, a single chemical sensor cannot localize this material and associate it with a person. We compensate this deficiency by fusing the output of multiple, distributed chemical sensors with kinematical data from laser-range scanners. Considering both the computer vision formation and the results of the person classification affords the localization of threats and a timely reaction of the security personnel.
Als rohstoffarme und exportorientierte Wirtschaftsnation ist die Bundesrepublik in ho- hem Maß auf die Sicherung und Sicherheit der Logistikketten im grenzüberschreiten- den Verkehr angewiesen. Angesichts der komplexen Transportstrukturen bei grenz- überschreitenden Transporten kommt den eingesetzten Kontroll- und Prüfverfahren besondere Bedeutung zu: Einerseits müssen Kostenbelastungen, Unterbrechungen und Verzögerungen in der Transportkette minimiert, andererseits besonders illegale Einfuhren, Transporte und Substanzen unterbunden werden. Von besonderer Bedeu- tung für Verdachts- bzw. Stichprobenkontrollen ist der Einsatz speziell trainierter Spür- hunde. Als besonders leistungsfähige ‚lebende Sensoren‘ sind sie in der Lage, eine Vielzahl von Stoffen zu detektieren. Der Einsatz von Spürhunden unterliegt allerdings engen Grenzen: Hoher Trainingsaufwand, eng begrenzte Einsatzdauer, begrenzte Verfügbarkeit. Die Entwicklung neuer, optimierter Einsatzverfahren für Spürhunde z. B. mit höheren Durchsatzraten und überprüfbarer Verlässlichkeit durch Einbindung technischer Systeme ist daher ein wichtiger Beitrag für die Sicherung und Sicherheit der Logistikketten.
Unter dem Begriff „Additive Fertigung“ werden alle Verfahren zusammengefasst, die dazu dienen Formteile aufgrund von CAD-Daten schichtweise aufzubauen. Dabei geschieht der Aufbau stets selektiv, entsprechend der durch die CAD-Daten vorgegebenen Positionen. Während für Metalle und Kunststoffe diese Technik bereits in der industriellen Anwendung etabliert ist, befindet sie sich im Bereich der keramischen Werkstoffe noch in einer frühen Entwicklungs- bzw. Anwendungsphase. Um so wichtiger ist es, den aktuellen Stand der Technik und das Potenzial für keramische Bauteile umfassend darzustellen.
Embryonic stem cells (ES) have the potential of long-term viability, selfrenewal and pluripotency which makes them interesting candidates for tissue engineering and gene therapy applications. On the other hand ethical and political issues arise while using theses cells and severe problems such as their tumorgenicity have not been solved yet. In the last couple of month a new source of cells with stem cell character was developed, the induced pluripotent stem cells (iPS). These cells are derived from differentiated adult cells via transduction of three transcription factors and show features similar to embryonic stem cells. Unfortunately, this includes the tumorgenicity which is even higher in those cells since the transcription factor transduction needed until now, is performed with retrovial vectors, which have a tumor potential on their own. Thus, adult stem cells are investigated extensively as alternative source of self-renewing cells. Human mesenchymal stem cells (HMSCs), which have in addition the advantage of potential autologous transplantation, can be found in various differentiated tissues since they are needed for maintenance and repair. They can be differentiated in chondrogenic, osteogenic, adipogenic and myogenic lineages which makes them an excellent tool for future tissue replacement strategies.
Advanced thermal gradient mechanical fatigue testing of CMSX-4 with an oxidation protection coating

In der vorliegenden Arbeit wird ein neuartiges Verfahren zur Echtzeitüberwachung von Laserbohrprozessen vorgestellt. Die Untersuchungen werden an unterschiedlichen Materialien unter Einsatz eines passiv-gütegeschalteten Nd:YAG Lasers durchgeführt. Prozessbegleitend findet eine Aufzeichnung der akustischen Emissionen mit anschließender Analyse durch schnelle Fourier-Transformation statt. Hierdurch lassen sich der Durchbruch beim Bohren durch ein Material sowie der Materialübergang mehrschichtiger Systeme detektieren. Die akustischen Messungen werden durchAuswertung der Pulsfolge des Lasers mittels einer Fotodiode gestützt. Hierbei zeigt sich eine gute Übereinstimmung der im akustischen Spektrum dominanten Frequenz mit der jeweils im Laserburstauftretenden Pulsfrequenz. Das vorgestellte Verfahren ermöglicht eine Echtzeitüberwachung beim Laserbohren mittels kostengünstiger und einfacher Hardware. Zudem zeichnet es sich im Gegensatz zu bestehenden Verfahren durch eine hohe Robustheit gegen äußere Störeinflüsse aus, da eine frequenzbasierte Auswertung stattfindet.
The human enzymes GLYAT (glycine N-acyltransferase), GLYATL1 (glutamine N-phenylacetyltransferase) and GLYATL2 (glycine N-acyltransferase-like protein 2) are not only important in the detoxification of xenobiotics via the human liver, but are also involved in the elimination of acyl residues that accumulate in the form of their coenzyme A (coA) esters in some rare inborn errors of metabolism. This concerns, for example, disorders in the degradation of branched-chain amino acids, such as isovaleric acidemia or propionic acidemia. In addition, they also assist in the elimination of ammonium, which is produced during the transamination of amino acids and accumulates in urea cycle defects. Sequence variants of the enzymes have also been investigated, which may provide evidence of impaired enzyme activities, from which therapy adjustments can potentially be derived. A modified Escherichia coli strain was chosen for the overexpression and partial biochemical characterization of the enzymes, which may allow solubility and proper folding. Since post-translational protein modifications are very limited in bacteria, we also attempted to overexpress the enzymes in HEK293 cells (human-derived). In addition to characterization via immunoblots and activity assays, intracellular localization of the enzymes was also performed using GFP coupling and confocal laser scanning microscopy in transfected HEK293 cells. The GLYATL2 enzyme may have tasks beyond detoxification and metabolic defects and the preliminary molecular biology work has been performed as part of this project - the enzyme activity determinations were outsourced to a co-supervised bachelor thesis. The enzyme activity determinations with purified recombinant human enzyme from Escherichia coli provided a threefold higher activity of the sequence variant p.(Asn156Ser) for GLYAT, which should be considered as the probably authentic wild type of the enzyme. In addition, a reduced activity of the GLYAT variant p.(Gln61Leu), which is very common in South Africa, was shown, which could be of particular importance in the treatment of isovaleric acidemia, which is also common in South Africa. Intracellularly, GLYAT and GLYATL1 could be localized mitochondrially. As the analyses have shown, sequence variations of GLYAT and GLYATL1 influence their enzyme activity. As an example, the GLYAT variant p.(Gln61Leu) is frequently found in South Africa. In the case of reduced GLYAT activity, patients could be increasingly treated with L-carnitine in the sense of an individualized therapy, since the conjugation of the toxic isovaleryl-coA with glycine is restricted by the GLYAT sequence variation. Activity-reducing variants identified in this project are of particular interest, as they may influence the treatment of certain metabolic defects.
Analysis of Synthetic Polymers and Copolymers by Pyrolysis- Gas Chromatography/Mass Spectrometry

Structural analysis and the study of degradation properties are important in order to understand and improve performance characteristics of synthetic polymers and copolymers in many industrial applications. Polymers/copolymers are inherently difficult to analyze because of their high molecular weight and lack of volatility. Analytical pyrolysis deals with the structural identification and quantitation of pyrolysis products with the ultimate aim of establishing the identity of the original material and the mechanisms of its thermal decomposition. The pyrolytic process is carried out in a pyrolyzer interfaced with analytical instrumentation such as gas chromatography (GC), mass spectrometry (MS), gas chromatography coupled with mass spectrometry (GC/MS), or with Fourier-transform infrared spectroscopy (GC/FTIR). By measurement and identification of pyrolysis products, the molecular composition of the original sample can often be reconstructed.This book is the outcome of contributions by experts in the field of pyrolysis and includes applications of the analytical pyrolysis-GC/MS to characterize the structure of synthetic organic polymers and lignocellulosic materials as well as cellulosic pulps and isolated lignins, solid wood, waste particle board, and bio-oil. Die Wiederverwertung von Kunststoffen (Kunststoffrecycling) kann in die werkstoffliche (materielle), die rohstoffliche (chemische) und die energetische Verwertung unterteilt werden. Beim werkstofflichen Kunststoffrecycling werden sortenreine Kunststoffreste gewaschen, gemahlen und von der Kunststoff verarbeitenden Industrie als Rohmaterial eingesetzt. Der chemische Aufbau des erhaltenen Werkstoffs (Re-Granulats) bleibt erhalten. Bei der rohstofflichen Verwertung werden Kunststoffreste zu Monomeren zurückgeführt. Die erhaltenen Monomere werden dann bei der Herstellung neuer Kunststoffe verwendet. Bei der energetischen Verwertung werden die Kunststoffreste der Zement- oder Stahlindustrie als Energieträger zugeführt.
Due to the use of fossil fuel resources, many environmental problems have been increasingly growing. Thus, the recent research focuses on the use of environment friendly materials from sustainable feedstocks for future fuels, chemicals, fibers and polymers. Lignocellulosic biomass has become the raw material of choice for these new materials. Recently, the research has focused on using lignin as a substitute material in many industrial applications. The antiradical and antimicrobial activity of lignin and lignin-based films are both of great interest for applications such as food packaging additives. DPPH assay was used to determine the antioxidant activity of Kraft lignin compared to Organosolv lignins from different biomasses. The purification procedure of Kraft lignin showed that double-fold selective extraction is the most efficient confirmed by UV-Vis, FTIR, HSQC, 31PNMR, SEC, and XRD. The antioxidant capacity was discussed regarding the biomass source, pulping process, and degree of purification. Lignin obtained from industrial black liquor are compared with beech wood samples: Biomass source influences the DPPH inhibition (softwood > grass) and the TPC (softwood < grass). DPPH inhibition affected by the polarity of the extraction solvent. Following the trend: ethanol > diethylether > acetone. Reduced polydispersity has positive influence on the DPPH inhibition. Storage decreased the DPPH inhibition but increased the TPC values. The DPPH assay was also used to discuss the antiradical activity of HPMC/lignin and HPMC/lignin/chitosan films. In both binary (HPMC/lignin) and ternary (HPMC/lignin/chitosan) systems the 5% addition showed the highest activity and the highest addition had the lowest. Both scavenging activity and antimicrobial activity are dependent on the biomass source; Organosolv of softwood > Kraft of softwood > Organosolv of grass. Lignins and lignin-containing films showed high antimicrobial activities against Gram-positive and Gram-negative bacteria at 35 °C and at low temperatures (0-7 °C). Purification of Kraft lignin has a negative effect on the antimicrobial activity while storage has positive effect. The lignin leaching in the produced films affected the activity positively and the chitosan addition enhances the activity for both Gram-positive and Gram-negative bacteria. Testing the films against food spoilage bacteria that grow at low temperatures revealed the activity of the 30% addition on HPMC/L1 film against both B. thermosphacta and P. fluorescens while L5 was active only against B. thermosphacta. In HPMC/lignin/chitosan films, the 5% addition exhibited activity against both food spoilage bacteria.
A soluble form of the complement receptor CD21 (sCD21) is shed from the lymphocyte surface. The sCD21 is able to bind all known ligands such as CD23, sCD23, Epstein-Barr virus and C3d in immune complexes. Here, we show the serum levels of sCD21 in sera the of antiphospholipid syndrome (APS) patients. Antiphospholipid syndrome is an autoimmune disorder in which autoantibodies cause heart attack, stroke and miscarriage. Antiphospholipid syndrome may appear as primary or in association with systemic lupus erythromatosus (SLE) and other autoimmune diseases. Here, we ask whether APS patients have different sCD21 titers compared to healthy persons and whether sCD21 levels correlate with the presence of anti-β2-GPI autoantibodies. We show that autoimmune APS patients have significantly reduced amounts of sCD21 in their sera, irrespective of the presence of anti-β2-GPI autoantibodies. In our APS patients cohort additional SLE, vasculities, DVT (deep vein thrombosis), fetal loss or thrombosis did not correlate to the reduced level of sCD21.
After replanting apple (Malus domestica Borkh.) on the same site severe growth suppressions, and a decline in yield and fruit quality are observed in all apple producing areas worldwide. The causes of this complex phenomenon, called apple replant disease (ARD), are only poorly understood up to now which is in part due to inconsistencies in terms and methodologies. Therefore we suggest the following definition for ARD: ARD describes a harmfully disturbed physiological and morphological reaction of apple plants to soils that faced alterations in their (micro-) biome due to the previous apple cultures. The underlying interactions likely have multiple causes that extend beyond common analytical tools in microbial ecology. They are influenced by soil properties, faunal vectors, and trophic cascades, with genotype-specific effects on plant secondary metabolism, particularly phytoalexin biosynthesis. Yet, emerging tools allow to unravel the soil and rhizosphere (micro-) biome, to characterize alterations of habitat quality, and to decipher the plant reactions. Thereby, deep insights into the reactions taking place at the root rhizosphere interface will be gained. Counteractions are suggested, taking into account that culture management should emphasize on improving soil microbial and faunal diversity as well as habitat quality rather than focus on soil disinfection.

Pyrolysis involves thermal fragmentation of the analytical sample at temperatures of 500–1400 °C. In the presence of an inert gas, reproducible decomposition products characteristic for the original polymer or copolymer sample are formed. The pyrolysis products are chromatographically separated using a fused-silica capillary column and are subsequently identified by interpretation of the obtained mass spectra or by using mass spectra libraries. The analytical technique eliminates the need for pretreatment by performing analyses directly on the solid or liquid polymer sample. In this article, application examples of analytical pyrolysis hyphenated to GC–MS for the identification of different polymeric materials in the plastic and automotive industry, dentistry, and occupational safety are demonstrated. For the first time, results of identification of commercial light-curing dental filling material and a car wrapping foil by pyrolysis–GC–MS are presented.

Gas chromatography with flame-ionization detection (FID) and gas chromatography-mass spectrometry (GC/MS) has been used for structure elucidation of long-chain primary n-alkyl amines after derivatization with trifluoroacetic anhydride (TFAA). Electron impact ionization- (EI) and positive chemical ionization- (PCI) mass spectra of trifluoroacetylated derivatives of the identified nalkyl amines are presented. The corrosion inhibiting n-alkyl amines were applied in the investigation of a new anticorrosive and antifouling formulation for water-steam circuit of energy systems in the power industry. The presented results are part of an EU-funded international collaboration with partners from research institutes and industry from Poland, Lithuania, Romania, France and Germany (EUREKA project BOILTREAT E!2426).
