Cathepsin K (CatK) is a target for the treatment of osteoporosis, arthritis, and bone metastasis. Peptidomimetics with a cyanohydrazide warhead represent a new class of highly potent CatK inhibitors; however, their binding mechanism is unknown. We investigated two model cyanohydrazide inhibitors with differently positioned warheads: an azadipeptide nitrile Gü1303 and a 3-cyano-3-aza-β-amino acid Gü2602. Crystal structures of their covalent complexes were determined with mature CatK as well as a zymogen-like activation intermediate of CatK. Binding mode analysis, together with quantum chemical calculations, revealed that the extraordinary picomolar potency of Gü2602 is entropically favoured by its conformational flexibility at the nonprimed-primed subsites boundary. Furthermore, we demonstrated by live cell imaging that cyanohydrazides effectively target mature CatK in osteosarcoma cells. Cyanohydrazides also suppressed the maturation of CatK by inhibiting the autoactivation of the CatK zymogen. Our results provide structural insights for the rational design of cyanohydrazide inhibitors of CatK as potential drugs.
Reliable and regional differentiated power forecasts are required to guarantee an efficient and economic energy transition towards renewable energies. Amongst other renewable energy technologies, e.g. wind mills, photovoltaic systems are an essential component of this transition being cost-efficient and simply to install. Reliable power forecasts are however required for a grid integration of photovoltaic systems, which among other data requires high-resolution spatio-temporal global irradiance data. Hence the generation of robust reviewed global irradiance data is an essential contribution for the energy transition.

Turbulent compressible flows are traditionally simulated using explicit time integrators applied to discretized versions of the Navier-Stokes equations. However, the associated Courant-Friedrichs-Lewy condition severely restricts the maximum time-step size. Exploiting the Lagrangian nature of the Boltzmann equation’s material derivative, we now introduce a feasible three-dimensional semi-Lagrangian lattice Boltzmann method (SLLBM), which circumvents this restriction. While many lattice Boltzmann methods for compressible flows were restricted to two dimensions due to the enormous number of discrete velocities in three dimensions, the SLLBM uses only 45 discrete velocities. Based on compressible Taylor-Green vortex simulations we show that the new method accurately captures shocks or shocklets as well as turbulence in 3D without utilizing additional filtering or stabilizing techniques other than the filtering introduced by the interpolation, even when the time-step sizes are up to two orders of magnitude larger compared to simulations in the literature. Our new method therefore enables researchers to study compressible turbulent flows by a fully explicit scheme, whose range of admissible time-step sizes is dictated by physics rather than spatial discretization.
Turbulent compressible flows are traditionally simulated using explicit Eulerian time integration applied to the Navier-Stokes equations. However, the associated Courant-Friedrichs-Lewy condition severely restricts the maximum time step size. Exploiting the Lagrangian nature of the Boltzmann equation's material derivative, we now introduce a feasible three-dimensional semi-Lagrangian lattice Boltzmann method (SLLBM), which elegantly circumvents this restriction. Previous lattice Boltzmann methods for compressible flows were mostly restricted to two dimensions due to the enormous number of discrete velocities needed in three dimensions. In contrast, this Rapid Communication demonstrates how cubature rules enhance the SLLBM to yield a three-dimensional velocity set with only 45 discrete velocities. Based on simulations of a compressible Taylor-Green vortex we show that the new method accurately captures shocks or shocklets as well as turbulence in 3D without utilizing additional filtering or stabilizing techniques, even when the time step sizes are up to two orders of magnitude larger compared to simulations in the literature. Our new method therefore enables researchers for the first time to study compressible turbulent flows by a fully explicit scheme, whose range of admissible time step sizes is only dictated by physics, while being decoupled from the spatial discretization.
Pursuant to Sustainable Development Goal (SDG) 15 of the 2030 Agenda for Sustainable Development of the United Nations, one pivotal target is to halt biodiversity loss. This paper’s objective is to analyze why and how German farmers hesitate to implement more than the prescriptive measures with regard to cross compliance and direct payments under the European Common Agricultural Policy (CAP) and what their aspirations are for possible incentives to bring biodiversity into focus. By applying a mixed methods approach, we investigate the experience of individual farmers by means of a qualitative approach followed by a quantitative study. This analysis sheds light on how farmers perceive indirect influencing factors and how these factors play a non-negligible role in farmers´ commitment to biodiversity. Economy, policy and society are intertwined and need to be considered from a multi-faceted perspective. In addition, an in-depth analysis is conducted based on online focus group discussions to determine whether farmers accept financial support, focusing on both action- and success-oriented payments. Our results highlight the importance of paying attention to the heterogeneity of farmers, their locations and, consequently, farmers’ different views on indirect drivers influencing agricultural processes, showing the complexity of the problem. Although farmers’ expectations can be met with financial allocations, other aspects must also be taken into account.
Modern Monte-Carlo-based rendering systems still suffer from the computational complexity involved in the generation of noise-free images, making it challenging to synthesize interactive previews. We present a framework suited for rendering such previews of static scenes using a caching technique that builds upon a linkless octree. Our approach allows for memory-efficient storage and constant-time lookup to cache diffuse illumination at multiple hitpoints along the traced paths. Non-diffuse surfaces are dealt with in a hybrid way in order to reconstruct view-dependent illumination while maintaining interactive frame rates. By evaluating the visual fidelity against ground truth sequences and by benchmarking, we show that our approach compares well to low-noise path-traced results, but with a greatly reduced computational complexity, allowing for interactive frame rates. This way, our caching technique provides a useful tool for global illumination previews and multi-view rendering.

Hand speed is particularly important in boxing both for protection against incoming blows and delivering blows. Sixteen amateur boxers (10 male, 6 female) with varying levels of experience from a boxing gym performed 20 jabs and 20 cross punches in air. The movement was recorded with a small wrist mounted accelerometer under the glove. The maximum velocity of each punch was determined from the RMS acceleration profile. The mean values of the jab maximal velocity was higher than the cross maximal velocity for 9 participants. The cross showed some dependence on reach (Spearman's correlation coefficient r = 0.57) and the jab dependence on experience (Spearman's correlation coefficient r = 0.56). The accelerometer technique has some promise for routine assessment of fist speed.

Isolated methylmalonic acidaemia (MMA) and propionic acidaemia (PA) are rare inherited metabolic diseases. Six years ago, a detailed evaluation of the available evidence on diagnosis and management of these disorders has been published for the first time. The article received considerable attention, illustrating the importance of an expert panel to evaluate and compile recommendations to guide rare disease patient care. Since that time, a growing body of evidence on transplant outcomes in MMA and PA patients and use of precursor free amino acid mixtures allows for updates of the guidelines. In this article, we aim to incorporate this newly published knowledge and provide a revised version of the guidelines. The analysis was performed by a panel of multidisciplinary health care experts, who followed an updated guideline development methodology (GRADE). Hence, the full body of evidence up until autumn 2019 was re‐evaluated, analysed and graded. As a result, 21 updated recommendations were compiled in a more concise paper with a focus on the existing evidence to enable well‐informed decisions in the context of MMA and PA patient care.

Green infrastructure improves environmental health in cities, benefits human health, and provides habitat for wildlife. Increasing urbanization has demanded the expansion of urban areas and transformation of existing cities. The adoption of compact design in urban planning is a recommended strategy to minimize environmental impacts; however, it may undermine green infrastructure networks within cities as it sets a battleground for urban space. Under this scenario, multifunctionality of green spaces is highly desirable but reconciling human needs and biodiversity conservation in a limited space is still a challenge. Through a systematic review, we first compiled urban green space's characteristics that affect mental health and urban wildlife support, and then identified potential synergies and trade-offs between these dimensions. A framework based on the One Health approach is proposed, synthesizing the interlinkages between green space quality, mental health, and wildlife support; providing a new holistic perspective on the topic. Looking at the human-wildlife-environment relationships simultaneously may contribute to practical guidance on more effective green space design and management that benefit all dimensions.
Motion capture, often abbreviated mocap, generally aims at recording any kind of motion -- be it from a person or an object -- and to transform it to a computer-readable format. Especially the data recorded from (professional and non-professional) human actors are typically used for analysis in e.g. medicine, sport sciences, or biomechanics for evaluation of human motion across various factors. Motion capture is also widely used in the entertainment industry: In video games and films realistic motion sequences and animations are generated through data-driven motion synthesis based on recorded motion (capture) data.
Although the amount of publicly available full-body-motion capture data is growing, the research community still lacks a comparable corpus of specialty motion data such as, e.g. prehensile movements for everyday actions. On the one hand, such data can be used to enrich (hand-over animation) full-body motion capture data - usually captured without hand motion data due to the drastic dimensional difference in articulation detail. On the other hand, it provides means to classify and analyse prehensile movements with or without respect to the concrete object manipulated and to transfer the acquired knowledge to other fields of research (e.g. from 'pure' motion analysis to robotics or biomechanics).
Therefore, the objective of this motion capture database is to provide well-documented, free motion capture data for research purposes.
The presented database GraspDB14 in sum contains over 2000 prehensile movements of ten different non-professional actors interacting with 15 different objects. Each grasp was realised five times by each actor. The motions are systematically named containing an (anonymous) identifier for each actor as well as one for the object grasped or interacted with.
The data were recorded as joint angles (and raw 8-bit sensor data) which can be transformed into positional 3D data (3D trajectories of each joint).
In this document, we provide a detailed description on the GraspDB14-database as well as on its creation (for reproducibility).
Chapter 2 gives a brief overview of motion capture techniques, freely available motion capture databases for both, full body motions and hand motions, and a short section on how such data is made useful and re-used. Chapter 3 describes the database recording process and details the recording setup and the recorded scenarios. It includes a list of objects and performed types of interaction. Chapter 4 covers used file formats, contents, and naming patterns. We provide various tools for parsing, conversion, and visualisation of the recorded motion sequences and document their usage in chapter 5.
The automated annotation of data from high throughput sequencing and genomics experiments is a significant challenge for bioinformatics. Most current approaches rely on sequential pipelines of gene finding and gene function prediction methods that annotate a gene with information from different reference data sources. Each function prediction method contributes evidence supporting a functional assignment. Such approaches generally ignore the links between the information in the reference datasets. These links, however, are valuable for assessing the plausibility of a function assignment and can be used to evaluate the confidence in a prediction. We are working towards a novel annotation system that uses the network of information supporting the function assignment to enrich the annotation process for use by expert curators and predicting the function of previously unannotated genes. In this paper we describe our success in the first stages of this development. We present the data integration steps that are needed to create the core database of integrated reference databases (UniProt, PFAM, PDB, GO and the pathway database Ara-Cyc) which has been established in the ONDEX data integration system. We also present a comparison between different methods for integration of GO terms as part of the function assignment pipeline and discuss the consequences of this analysis for improving the accuracy of gene function annotation. The methods and algorithms presented in this publication are an integral part of the ONDEX system which is freely available from http://ondex.sf.net/.
Target meaning representations for semantic parsing tasks are often based on programming or query languages, such as SQL, and can be formalized by a context-free grammar. Assuming a priori knowledge of the target domain, such grammars can be exploited to enforce syntactical constraints when predicting logical forms. To that end, we assess how syntactical parsers can be integrated into modern encoder-decoder frameworks. Specifically, we implement an attentional SEQ2SEQ model that uses an LR parser to maintain syntactically valid sequences throughout the decoding procedure. Compared to other approaches to grammar-guided decoding that modify the underlying neural network architecture or attempt to derive full parse trees, our approach is conceptually simpler, adds less computational overhead during inference and integrates seamlessly with current SEQ2SEQ frameworks. We present preliminary evaluation results against a recurrent SEQ2SEQ baseline on GEOQUERY and ATIS and demonstrate improved performance while enforcing grammatical constraints.
With the debates on climate change and sustainability, a reduction of the share of cars in the modal split has become increasingly prevalent in both public and academic discourse. Besides some motivational approaches, there is a lack of ICT artifacts that successfully raise the ability of consumers to adopt sustainable mobility patterns. To further understand the requirements and the design of these artifacts within everyday mobility adopted a practice-lens. This lens is helpful to get a broader perspective on the use of ICT artifacts along consumers’ transformational journey towards sustainable mobility practices. Based on 12 retrospective interviews with car-free mobility consumers, we argue that artifacts should not be viewed as ’magic-bullet’ solutions but should accompany the complex transformation of practices in multifaceted ways. Moreover, we highlight in particular the difficulties of appropriating shared infrastructures and aligning own practices with them. This opens up a design space to provide more support for these kinds of material-interactions, to provide access to consumption infrastructures and make them usable, rather than leaving consumers alone with increased motivation.
The year 2020 was dominated by the Corona pandemic, which abruptly changed everyday life at the university. Bonn-Rhein-Sieg University of Applied Sciences (H-BRS) succeeded not only in maintaining operations under these difficult circumstances, but also in enriching them with new ideas and insights. This difficult year 2020 has opened up new perspectives, especially for teaching, but also for research and administration. This annual report, to which we have given the motto "Walking - knowing the point of view, taking direction, showing attitude", tells of this. It shows which paths the H-BRS has taken in the past year. Some routes were rocky, and some slopes could only be overcome with combined efforts. Often, however, new terrain was opened up. The aim was always to find answers to the multi-layered, complex questions of our time - whether with regard to digitalisation, climate change or the social responsibility of science.

Ghanaian tertiary graduates' perception of entrepreneurship education on employment opportunities
(2017)
This study focuses on whether entrepreneurship education increases entrepreneurial interest in students to set up new businesses. Entrepreneurship is a core course taken in the third year by all students of Ho Technical University. Out of the 1329 population of level 300 students of the 2016/2017 academic year, data were collected by convenience sampling from 325 (217 males and 108 females) with mean age of 24.75 years from 14 departments of four faculties. The students responded to 43-survey items derived from reviewed literature on a 5-Point Likert-Scale. It is concluded that more than 84% of the respondents agreed that entrepreneurship education informed students about entrepreneurship through the acquisition of practical skills, knowledge about acquisition of personal orientation, knowledge about business management principles and the availability of entrepreneurial support agencies. This shows that the students are highly confident of setting up their own businesses through the knowledge acquired. The study therefore has important implications for policy makers, management of tertiary institutions, students and educational evaluators on how to ensure that tertiary graduates set up entrepreneurship ventures in order to partially solve the unemployment problem in Ghana.

This paper examines how students learn to collaborate in English by participating in an intercultural project that focuses on teaching students to work together on a digital writing project using various online tools, and participated in this digital collaboration project. Mixed groups of students, two French and two German, used several synchronous and asynchronous tools to communicate with their counterparts (Facebook, WordPress blog, WIMS e-learning platform, email, videoconferencing). Students had to produce an article together, comparing French and German attitudes about a topic they negotiated freely in their groups. Before publishing their post, students were expected to peer-review the article written by their group. Once published, the stage consisted of voting for the best posts on the e-learning platform, WIMS. A videoconference was also organized to create cohesion between the participants. The result of the student evaluations, together with the administrative, technical vastly differing university setups is presented.
A principal step towards solving diverse perception problems is segmentation. Many algorithms benefit from initially partitioning input point clouds into objects and their parts. In accordance with cognitive sciences, segmentation goal may be formulated as to split point clouds into locally smooth convex areas, enclosed by sharp concave boundaries. This goal is based on purely geometrical considerations and does not incorporate any constraints, or semantics, of the scene and objects being segmented, which makes it very general and widely applicable. In this work we perform geometrical segmentation of point cloud data according to the stated goal. The data is mapped onto a graph and the task of graph partitioning is considered. We formulate an objective function and derive a discrete optimization problem based on it. Finding the globally optimal solution is an NP-complete problem; in order to circumvent this, spectral methods are applied. Two algorithms that implement the divisive hierarchical clustering scheme are proposed. They derive graph partition by analyzing the eigenvectors obtained through spectral relaxation. The specifics of our application domain are used to automatically introduce cannot-link constraints in the clustering problem. The algorithms function in completely unsupervised manner and make no assumptions about shapes of objects and structures that they segment. Three publicly available datasets with cluttered real-world scenes and an abundance of box-like, cylindrical, and free-form objects are used to demonstrate convincing performance. Preliminary results of this thesis have been contributed to the International Conference on Autonomous Intelligent Systems (IAS-13).
Apple replant disease (ARD) is a soil-borne disease, which is of particular importance for fruit tree nurseries and fruit growers. The disease manifests by a poor vegetative development, stunted growth, and reduced yield in terms of quantity and quality, if apple plants (usually rootstocks) are replanted several times at the same site. Genotype-specific differences in the reaction of apple plants to ARD are documented, but less is known about the genetic mechanisms behind this symptomatology. Recent transcriptome analyses resulted in a number of candidate genes possibly involved in the plant response. In the present study, the expression of 108 selected candidate genes was investigated in root and leaf tissue of four different apple genotypes grown in untreated ARD soil and ARD soil disinfected by γ-irradiation originating from two different sites in Germany. Thirty-nine out of the 108 candidate genes were differentially expressed in roots by taking a p-value of < 0.05 and a fold change of > 1.5 as cutoff. Sixteen genes were more than 4.5-fold upregulated in roots of plants grown in ARD soil. The four genes MNL2 (putative mannosidase); ALF5 (multi antimicrobial extrusion protein); UGT73B4 (uridine diphosphate (UDP)-glycosyltransferase 73B4), and ECHI (chitin-binding) were significantly upregulated in roots. These genes seem to be related to the host plant response to ARD, although they have never been described in this context before. Six of the highly upregulated genes belong to the phytoalexin biosynthesis pathway. Their genotype-specific gene expression pattern was consistent with the phytoalexin content measured in roots. The biphenyl synthase (BIS) genes were found to be useful as early biomarkers for ARD, because their expression pattern correlated well with the phenotypic reaction of the Malus genotypes investigated.
High-dimensional and multi-variate data from dynamical systems such as turbulent flows and wind turbines can be analyzed with deep learning due to its capacity to learn representations in lower-dimensional manifolds. Two challenges of interest arise from data generated from these systems, namely, how to anticipate wind turbine failures and how to better understand air flow through car ventilation systems. There are deep neural network architectures that can project data into a lower-dimensional space with the goal of identifying and understanding patterns that are not distinguishable in the original dimensional space. Learning data representations in lower dimensions via non-linear mappings allows one to perform data compression, data clustering (for anomaly detection), data reconstruction and synthetic data generation.
In this work, we explore the potential that variational autoencoders (VAE) have to learn low-dimensional data representations in order to tackle the problems posed by the two dynamical systems mentioned above. A VAE is a neural network architecture that combines the mechanisms of the standard autoencoder and variational bayes. The goal here is to train a neural network to minimize a loss function defined by a reconstruction term together with a variational term defined as a Kulback-Leibler (KL) divergence.
The report discusses the results obtained for the two different data domains: wind turbine time series and turbulence data from computational fluid dynamics (CFD) simulations.
We report on the reconstruction, clustering and unsupervised anomaly detection of wind turbine multi-variate time series data using a variant of a VAE called Variational Recurrent Autoencoder (VRAE). We trained a VRAE to cluster normal and abnormal wind turbine series (two class problem) as well as normal and multiple abnormal series (multi-class problem). We found that the model is capable of distinguishing between normal and abnormal cases by reducing the dimensionality of the input data and projecting it to two dimensions using techniques such as Principal Component Analysis (PCA) and t-distributed stochastic neighbor embedding (t-SNE). A set of anomaly scoring methods is applied on top of these latent vectors in order to compute unsupervised clustering. We have achieved an accuracy of up to 96% with the KM eans + + algorithm.
We also report the data reconstruction and generation results of two dimensional turbulence slices corresponding to CFD simulation of a HVAC air duct. For this, we have trained a Convolutional Variational Autoencoder (CVAE). We have found that the model is capable of reconstructing laminar flows up to a certain degree of resolution as well generating synthetic turbulence data from the learned latent distribution.
In the field of automatic music generation, one of the greatest challenges is the consistent generation of pieces continuously perceived positively by the majority of the audience since there is no objective method to determine the quality of a musical composition. However, composing principles, which have been refined for millennia, have shaped the core characteristics of today's music. A hybrid music generation system, mlmusic, that incorporates various static, music-theory-based methods, as well as data-driven, subsystems, is implemented to automatically generate pieces considered acceptable by the average listener. Initially, a MIDI dataset, consisting of over 100 hand-picked pieces of various styles and complexities, is analysed using basic music theory principles, and the abstracted information is fed into explicitly constrained LSTM networks. For chord progressions, each individual network is specifically trained on a given sequence length, while phrases are created by consecutively predicting the notes' offset, pitch and duration. Using these outputs as a composition's foundation, additional musical elements, along with constrained recurrent rhythmic and tonal patterns, are statically generated. Although no survey regarding the pieces' reception could be carried out, the successful generation of numerous compositions of varying complexities suggests that the integration of these fundamentally distinctive approaches might lead to success in other branches.
Generating and visualizing large areas of vegetation that look natural makes terrain surfaces much more realistic. However, this is a challenging field in computer graphics, because ecological systems are complex and visually appealing plant models are geometrically detailed. This work presents Silva (System for the Instantiation of Large Vegetated Areas), a system to generate and visualize large vegetated areas based on the ecological surrounding. Silva generates vegetation on Wang-tiles with associated reusable distributions enabling multi-level instantiation. This paper presents a method to generate Poisson Disc Distributions (PDDs) with variable radii on Wang-tile sets (without a global optimization) that is able to generate seamless tilings. Because Silva has a freely configurable generation pipeline and can consider plant neighborhoods it is able to incorporate arbitrary abiotic and biotic components during generation. Based on multi-levelinstancing and nested kd-trees, the distributions on the Wang-tiles allow their acceleration structures to be reused during visualization. This enables Silva to visualize large vegetated areas of several hundred square kilometers with low render times and a small memory footprint.
Females are influenced more than males by visual cues during many spatial orientation tasks; but females rely more heavily on gravitational cues during visual-vestibular conflict. Are there gender biases in the relative contributions of vision, gravity and the internal representation of the body to the perception of upright? And might any such biases be affected by low gravity? 16 participants (8 female) viewed a highly polarized visual scene tilted ±112° while lying supine on the European Space Agency's short-arm human centrifuge. The centrifuge was rotated to simulate 24 logarithmically spaced g-levels along the long axis of the body (0.04-0.5g at ear-level). The perception of upright was measured using the Oriented Character Recognition Test (OCHART). OCHART uses the ambiguous symbol "p" shown in different orientations. Participants decided whether it was a "p" or a "d" from which the perceptual upright (PU) can be calculated for each visual/gravity combination. The relative contribution of vision, gravity and the internal representation of the body were then calculated. Experiments were repeated while upright. The relative contribution of vision on the PU was less in females compared to males (t=-18.48, p≤0.01). Females placed more emphasis on the gravity cue instead (f:28.4%, m:24.9%) while body weightings were constant (f:63.0%, m:63.2%). When upright (1g) in this and other studies (e.g., Barnett-Cowan et al. 2010, EJN, 31,1899) females placed more emphasis on vision in this task than males. The reduction in weight allocated by females to vision when in simulated low-gravity conditions compared to when upright under normal gravity may be related to similar female behaviour in response to other instances of visual-vestibular conflict. Why this is the case and at which point the perceptual change happens requires further research.

In thyroid carcinoma cells, the soluble βgalactosidespecific lectin, galectin3, is extra and intracellularly expressed and plays a significant role in thyroid cancer diagnosis. The functional relevance of this molecule, particularly in its extracellular environment however, warrants further elucidation. To gain insight into this topic, the present study characterized principal functional properties of galectin3 in 3 commonly used thyroid carcinoma cell lines (BCPAP, Cal62 and FTC133) that express the molecule intra and extracellulary. Cellintrinsic galectin3 harbors a functional carbohydrate recognition domain as determined by affinity purification. Moreover, cell surface expressed galectin3 can be partially removed by treatment with lactose or asialofetuin, but not with sucrose. Thyroid carcinoma cells adhere to substratebound galectin3 in a βgalactosidespecific manner, whereby only cell adhesion, but not cell migration is promoted. Thus, thyroid tumor cells harbor functional active galectin3 that, inter alia, specifically interacts with cell surfaceexpressed molecular ligands in a βgalactosidedependent manner, whereby the molecule can at least interfere with cell adhesion. The modulation of galectin3 expression level or its ligands in such tumor cells could be of therapeutic interest and needs further experimental clarification.

What does ‘desirable’ or ‘sustainable’ mean in the context of labour migration? And what should programmes geared towards making migration more compatible with development look like? These questions provided the starting point for the ‘Sustainable Labour Migration’ research project implemented by Hochschule Bonn-Rhein-Sieg University of Applied Sciences between December 2020 and August 2022. The project looked at how sustainability in different transnational skills partnership schemes was perceived by different stakeholders in three countries chosen as case studies: Georgia, Kosovo and Vietnam. Embracing the notion of a ‘triple win’, many transnational skills partnership schemes aim to deliver benefits for their main stakeholder groups. As well as reflecting critically on this triple-win narrative, this paper also argues for a more nuanced approach in order to grasp the complexity of skilled labour migration. The paper introduces one such approach, namely the sustainable labour migration framework, and highlights the key elements of the research project. It details the methodology used in the study (systematic literature review – employer survey – semi-structured, in-depth interviews – focus group interviews) and explores the perception of sustainability in skilled labour migration with a focus on the cost/benefit ratio, the relationship between vocational education and training and labour migration, and the various arrangements for the partnerships employed in the schemes. In introducing the key findings from the three countries selected for the case studies, the paper highlights that the perception of sustainability is not underpinned by a comprehensive understanding of the term amongst most stakeholders. Within all the schemes, however, some stakeholders identified elements which make a positive contribution to development in the respective countries of origin and thus identify elements of sustainable labour migration with room for improvement in multiple areas. The paper concludes with a presentation of overall policy recommendations: The ongoing reform of Germany’s labour migration policy should be accompanied by more development-oriented activities. As part of this, the German Federal Ministry for Economic Cooperation and Development (BMZ) should stand up more forcefully for the needs of migrants and potential partner countries in order to reduce existing inequalities
We are happy to present you the special issue on Best Practice in Robot Software Development of the Journal on Software Engineering for Robotics! The spark for this special issue came during the eighth workshop on Software Development and Integration in Robotics (SDIR) at the 2013 IEEE International Conference on Robotics and Automation. The workshop focused on Robot Software Architectures, and the fruitful discussions made it clear that the design, development, and deployment of robot software is always an interplay between competing aspects. These are often couched in antagonistic pairs, such as dependability versus performance, and prominently include quality attributes as well as functional, nonfunctional, and application requirements.

A Fourier scatterometry setup is evaluated to recover the key parameters of optical phase gratings. Based on these parameters, systematic errors in the printing process of two-photon polymerization (TPP) gray-scale lithography three-dimensional printers can be compensated, namely tilt and curvature deviations. The proposed setup is significantly cheaper than a confocal microscope, which is usually used to determine calibration parameters for compensation of the TPP printing process. The grating parameters recovered this way are compared to those obtained with a confocal microscope. A clear correlation between confocal and scatterometric measurements is first shown for structures containing either tilt or curvature. The correlation is also shown for structures containing a mixture of tilt and curvature errors (squared Pearson coefficient r2 = 0.92). This compensation method is demonstrated on a TPP printer: a diffractive optical element printed with correction parameters obtained from Fourier scatterometry shows a significant reduction in noise as compared to the uncompensated system. This verifies the successful reduction of tilt and curvature errors. Further improvements of the method are proposed, which may enable the measurements to become more precise than confocal measurements in the future, since scatterometry is not affected by the diffraction limit.
While Anglo-Saxon HEIs focus on a strong educational background and personal development of students, the German system, in particular Universities of Applied Sciences, emphasize employability through the transfer of job-related professional and soft skills. In this context, learning by practical application of skills has become an important instrument. Concepts for linking theory and application include research-based learning, practical internships or service learning – methods, which also maintain high standards of academic education.

Food losses occur for many reasons at all stages of supply chains for fruits, vegetables and potatoes. They cause immense economic, environmental and social costs – not only in developing countries but also in developed countries. According to the European Commission, about 90 million tonnes of food are wasted annually in Europe alone. However, particularly for the early stages of supply chains for fruits, vegetables and potatoes there is still a lack of reliable data. Thus, one objective of this study is to contribute to the quantification of food losses between field and retail, where the main focus is set on potatoes, apples, carrots, strawberries and asparagus. Furthermore, neither reasons why products are removed from the supply chains nor their alternative uses are fully examined yet. This is why, the study takes a look on those issues, too. Results are based on data from an online survey among producers of fruits, vegetables and potatoes in North-Rhine Westphalia, Germany and on interviews with producers and other supply chain experts. Findings suggest that the products’ size and form, their storage capabilities and food safety issues have big impacts on food losses. Despite a small sample size, these findings are in line with recent studies.
Background: Migration of mature and immature leukocytes in response to chemokines is not only essential during inflammation and host defense, but also during development of the hematopoietic system. Many molecules implicated in migratory polarity show uniform cellular distribution under non-activated conditions, but acquire a polarized localization upon exposure to migratory cues.
Methodology/Principal Findings: Here, we present evidence that raft-associated endocytic proteins (flotillins) are preassembled in lymphoid, myeloid and primitive hematopoietic cells and accumulate in the uropod during migration. Furthermore, flotillins display a polarized distribution during immunological synapse formation. Employing the membrane lipid-order sensitive probe Laurdan, we show that flotillin accumulation in the immunological synapse is concomittant with membrane ordering in these regions.
Conclusions: Together with the observation that flotillin polarization does not occur in other polarized cell types such as polarized epithelial cells, our results suggest a specific role for flotillins in hematopoietic cell polarization. Based on our results, we propose that in hematopoietic cells, flotillins provide intrinsic cues that govern segregation of certain microdomain-associated molecules during immune cell polarization.
The labor market is dynamic and frequently calls for new skills, knowledge, and abilities. The changing needs of industry place a higher demand on institutions of higher learning to monitor trends in labor needs, identify skill gaps, and to use industry insights for developing programs and curricula that mold human resources to create value for employers and society at large. While several institutions of higher learning are responsive to industry needs through curricula reviews and the development of new programs, little attention is given to pedagogical issues that affect the delivery of knowledge and the development of skills intended by various education programs. Consequently, teachers are entrusted with the freedom to decide the teaching methods that are appropriate under each circumstance. Despite the changing face of the labor market, not much energy has been channeled towards adjusting teaching methods for effective delivery of skills required by students. The failure to adjust teaching methods for training graduates has led to what is commonly known as ‘halfbaked graduates’. In other words, graduates who lack the skills and abilities necessary for placement in the industry. However, the success of an institution of higher learning is illustrated by its ability to train people who perfectly match the needs of the industry.
The visual and auditory quality of computer-mediated stimuli for virtual and extended reality (VR/XR) is rapidly improving. Still, it remains challenging to provide a fully embodied sensation and awareness of objects surrounding, approaching, or touching us in a 3D environment, though it can greatly aid task performance in a 3D user interface. For example, feedback can provide warning signals for potential collisions (e.g., bumping into an obstacle while navigating) or pinpointing areas where one’s attention should be directed to (e.g., points of interest or danger). These events inform our motor behaviour and are often associated with perception mechanisms associated with our so-called peripersonal and extrapersonal space models that relate our body to object distance, direction, and contact point/impact. We will discuss these references spaces to explain the role of different cues in our motor action responses that underlie 3D interaction tasks. However, providing proximity and collision cues can be challenging. Various full-body vibration systems have been developed that stimulate body parts other than the hands, but can have limitations in their applicability and feasibility due to their cost and effort to operate, as well as hygienic considerations associated with e.g., Covid-19. Informed by results of a prior study using low-frequencies for collision feedback, in this paper we look at an unobtrusive way to provide spatial, proximal and collision cues. Specifically, we assess the potential of foot sole stimulation to provide cues about object direction and relative distance, as well as collision direction and force of impact. Results indicate that in particular vibration-based stimuli could be useful within the frame of peripersonal and extrapersonal space perception that support 3DUI tasks. Current results favor the feedback combination of continuous vibrotactor cues for proximity, and bass-shaker cues for body collision. Results show that users could rather easily judge the different cues at a reasonably high granularity. This granularity may be sufficient to support common navigation tasks in a 3DUI.
In recent years, a plethora of observations with high spectral resolution of sub-millimetre and far-infrared transitions of methylidene (CH), conducted with Herschel and SOFIA, have demonstrated this radical to be a valuable proxy for molecular hydrogen that can be used for characterising molecular gas within the interstellar medium on a Galactic scale, including the CO-dark component. We report the discovery of the 13CH isotopologue in the interstellar medium using the upGREAT receiver on board SOFIA. We have detected the three hyperfine structure components of the ≈2 THz frequency transition from its X2Π1∕2 ground-state towards the high-mass star-forming regions Sgr B2(M), G34.26+0.15, W49(N), and W51E and determined 13CH column densities. The ubiquity of molecules containing carbon in the interstellar medium has turned the determination of the ratio between the abundances of the two stable isotopes of carbon, 12C/13C, into a cornerstone for Galactic chemical evolution studies. Whilst displaying a rising gradient with galactocentric distance, this ratio, when measured using observations of different molecules (CO, H2CO, and others), shows systematic variations depending on the tracer used. These observed inconsistencies may arise from optical depth effects, chemical fractionation, or isotope-selective photo-dissociation. Formed from C+ either through UV-driven or turbulence-driven chemistry, CH reflects the fractionation of C+, and does not show any significant fractionation effects, unlike other molecules that were previously used to determine the 12C/13C isotopic ratio. This makes it an ideal tracer for the 12C/13C ratio throughout the Galaxy. By comparing the derived column densities of 13CH with previously obtained SOFIA data of the corresponding transitions of the main isotopologue 12CH, we therefore derive 12C/13C isotopic ratios toward Sgr B2(M), G34.26+0.15, W49(N) and W51E. Adding our values derived from 12∕13CH to previous calculations of the Galactic isotopic gradient, we derive a revised value of 12C/13C = 5.87(0.45)RGC + 13.25(2.94).
A system that interacts with its environment can be much more robust if it is able to reason about the faults that occur in its environment, despite perfect functioning of its internal components. For robots, which interact with the same environment as human beings, this robustness can be obtained by incorporating human-like reasoning abilities in them. In this work we use naive physics to enable reasoning about external faults in robots. We propose an approach for diagnosing external faults that uses qualitative reasoning on naive physics concepts for diagnosis. These concepts are mainly individual properties of objects that define their state qualitatively. The reasoning process uses physical laws to generate possible states of the concerned object(s), which could result into a detected external fault. Since effective reasoning about any external fault requires the information of relevant properties and physical laws, we associate different properties and laws to different types of faults which can be detected by a robot. The underlying ontology of this association is proposed on the basis of studies conducted (by other researchers) on reasoning of physics novices about everyday physical phenomena. We also formalize some definitions of properties of objects into a small framework represented in First-Order logic. These definitions represent naive concepts behind the properties and are intended to be independent from objects and circumstances. The definitions in the framework illustrates our proposal of using different biased definitions of properties for different types of faults. In this work, we also present a brief review of important contributions in the area of naive/qualitative physics. These reviews help in understanding the limitations of naive/qualitative physics in general. We also apply our approach to simple scenarios to asses its limitations in particular. Since this work was done independent of any particular real robotic system, it can be seen as a theoretical proof of the concept of usefulness of naive physics for external fault reasoning in robotics.
Analytical redundancy relations are fundamental in model-based fault detection and isolation. Their numerical evaluation yields a residual that may serve as a fault indicator. Considering switching linear time-invariant system models that use ideal switches, it is shown that analytical redundancy relations can be systematically deduced from a diagnostic bond graph with fixed causalities that hold for all modes of operation. Moreover, as to a faultless system, the presented bond graph–based approach enables to deduce a unique implicit state equation with coefficients that are functions of the discrete switch states. Devices or phenomena with fast state transitions, for example, electronic diodes and transistors, clutches, or hard mechanical stops are often represented by ideal switches which give rise to variable causalities. However, in the presented approach, fixed causalities are assigned only once to a diagnostic bond graph. That is, causal strokes at switch ports in the diagnostic bond graph reflect only the switch-state configuration in a specific system mode. The actual discrete switch states are implicitly taken into account by the discrete values of the switch moduli. The presented approach starts from a diagnostic bond graph with fixed causalities and from a partitioning of the bond graph junction structure and systematically deduces a set of equations that determines the wanted residuals. Elimination steps result in analytical redundancy relations in which the states of the storage elements and the outputs of the ideal switches are unknowns. For the later two unknowns, the approach produces an implicit differential algebraic equations system. For illustration of the general matrix-based approach, an electromechanical system and two small electronic circuits are considered. Their equations are directly derived from a diagnostic bond graph by following causal paths and are reformulated so that they conform with the matrix equations obtained by the formal approach based on a partitioning of the bond graph junction structure. For one of the three mode-switching examples, a fault scenario has been simulated.
Extraction of text information from visual sources is an important component of many modern applications, for example, extracting the text from traffic signs on a road scene in an autonomous vehicle. For natural images or road scenes this is a unsolved problem. In this thesis the use of histogram of stroke widths (HSW) for character and noncharacter region classification is presented. Stroke widths are extracted using two methods. One is based on the Stroke Width Transform and another based on run lengths. The HSW is combined with two simple region features– aspect and occupancy ratios– and then a linear SVM is used as classifier. One advantage of our method over the state of the art is that it is script-independent and can also be used to verify detected text regions with the purpose of reducing false positives. Our experiments on generated datasets of Latin, CJK, Hiragana and Katakana characters show that the HSW is able to correctly classify at least 90% of the character regions, a similar figure is obtained for non-character regions. This performance is also obtained when training the HSW with one script and testing with a different one, and even when characters are rotated. On the English and Kannada portions of the Chars74K dataset we obtained over 95% correctly classified character regions. The use of raycasting for text line grouping is also proposed. By combining it with our HSW-based character classifier, a text detector based on Maximally Stable Extremal Regions (MSER) was implemented. The text detector was evaluated on our own dataset of road scenes from the German Autobahn, where 65% precision, 72% recall with a f-score of 69% was obtained. Using the HSW as a text verifier increases precision while slightly reducing recall. Our HSW feature allows the building of a script-independent and low parameter count classifier for character and non-character regions.
Advanced driver assistance systems (ADAS) are technology systems and devices designed as an aid to the driver of a vehicle. One of the critical components of any ADAS is the traffic sign recognition module. For this module to achieve real-time performance, some preprocessing of input images must be done, which consists of a traffic sign detection (TSD) algorithm to reduce the possible hypothesis space. Performance of TSD algorithm is critical.
One of the best algorithms used for TSD is the Radial Symmetry Detector (RSD), which can detect both Circular [7] and Polygonal traffic signs [5]. This algorithm runs in real-time on high end personal computers, but computational performance of must be improved in order to be able to run in real-time in embedded computer platforms.
To improve the computational performance of the RSD, we propose a multiscale approach and the removal of a gaussian smoothing filter used in this algorithm. We evaluate the performance on both computation times, detection and false positive rates on a synthetic image dataset and on the german traffic sign detection benchmark [29].
We observed significant speedups compared to the original algorithm. Our Improved Radial Symmetry Detector is up to 5.8 times faster than the original on detecting Circles, up to 3.8 times faster on Triangle detection, 2.9 times faster on Square detection and 2.4 times faster on Octagon detection. All of this measurements were observed with better detection and false positive rates than the original RSD.
When evaluated on the GTSDB, we observed smaller speedups, in the range of 1.6 to 2.3 times faster for Circle and Regular Polygon detection, but for Circle detection we observed a decreased detection rate than the original algorithm, while for Regular Polygon detection we always observed better detection rates. False positive rates were high, in the range of 80% to 90%.
We conclude that our Improved Radial Symmetry Detector is a significant improvement of the Radial Symmetry Detector, both for Circle and Regular polygon detection. We expect that our improved algorithm will lead the way to obtain real-time traffic sign detection and recognition in embedded computer platforms.
This paper aims to assess farmers’ challenges in enhancing biodiversity. The so-called “trilemma” (WBGU 2021) of land use stems from the multiple demands made on land for the benefit of mitigating climate change, securing food, and maintaining biodiversity. Agriculture is accused of maladministration, causing soil contamination, animal cruelty, bee mortality, and climate change. However, farmers play a key role in overcoming upcoming sustainability challenges. While their supportive role is urgently needed, farmers find themselves caught between a “rock” and a ”hard place”. Consumers call for sustainable production and affordable food products without pesticide residues, demanding enough for all. Farmers are restricted by the wants and needs of consumers who are influenced by interest groups and exposed to interdependent direct and indirect influencing factors. They need to balance the scrutiny of the critical public as well as the regulatory control. In this paper, we collected and surveyed the data of farmers within or close to the 21 selected nature protected areas of the DINA (Diversity of Insects in Nature protected Areas) Project, using a mixed methods approach with a semi-structured questionnaire considering issues’ interdependencies and the complexity of today´s problems. The conflicts and obstacles faced by farmers were assessed. The results reflect the farmers’ willingness and the importance of receiving appreciation for implementing biodiversity measures. These results, complemented by a following quantitative study, are the basis for recommendations for policymakers and farmers in all German nature protected areas.
New sustainable, environmentally friendly materials for thermal insulation of buildings are necessary to reduce their carbon footprints. In this study, Miscanthus fiber-reinforced geopolymer composites, foamed with sodium dodecyl sulfate (SDS), were developed using fly ash as a geopolymer precursor. The effects of fiber content, fiber size, curing temperature, foaming agent content, fumed silica specific surface area and fumed silica content on thermal conductivity and compressive strength were evaluated using a Plackett-Burman design of experiment. Furthermore, the microstructure of geopolymer composites was investigated using X-ray diffraction (XRD), X-ray micro-computed tomography (μCT) and scanning electron microscopy (SEM). The measured characteristic values were in the following ranges: Thermal conductivity 0.057 W (m K)−1 to 0.127 W (m K)−1, compressive strength 0.007 MPa–0.719 MPa and porosity 49 vol% to 76 vol%. The results reveal an enhancement of thermal conductivity by elevated fiber size and foaming agent content. In contrast, the compressive strength is enhanced by high fiber content. Additionally, SEM images indicate a good interaction between the fibers and the geopolymer matrix, because nearly the whole fiber surface is covered by the geopolymer.
