Unkonventionelle Spreng- und Brandvorrichtungen sind Bedrohungen in den weltweiten Konfliktherden und werden bei terroristischen Aktivitäten verwendet. Der Schutz von Menschen und Material erfordert daher effektive Gegenmaßnahmen. Dazu gehört auch die Anforderung an Sicherheitskräfte oder militärisches Personal, unbekannte Substanzfunde mit geringem zeitlichem und logistischem Aufwand vor Ort als gefährdend oder unkritisch einzustufen. Um Explosivstoffe von nicht-explosiven Materialien zu unterscheiden, kann die bei Explosivstoffen initiierbare, stark exotherme Reaktion genutzt werden. Diese resultiert in Strahlungsemissionen sowie in lokaler Druck- und Temperaturerhöhung. Die Messung dieser Reaktionseffekte und die Anforderung an eine mobile, einfach zu bedienende und robuste Analytik werden durch ein System ermöglicht, das Proben im einstelligen mg-Bereich durch schnelles Erhitzen auf mikrostrukturierten Heizern zum chemischen Umsatz anregt. Die emittierte Strahlung wird mit Photodioden im Bereich des sichtbaren und nah-infraroten Lichts aufgenommen, ein Sensor registriert die Druckerhöhung in einer geschlossenen Versuchskammer. In einem zweiten Aufbau werden die gasförmigen Reaktionsprodukte über ein Sensorarray von vier kommerziellen Gassensoren geleitet und die Signalantworten der Halbleitergassensoren mittels Hauptkomponentenanalyse ausgewertet. Die Ergebnisse zeigen, dass die schnelle thermische Aktivierung für die untersuchten primären Explosivstoffe, Treibladungspulver, sowie Trinitrotoluol (TNT) reproduzierbar erfolgt. Nicht-Explosivstoffe werden dabei im untersuchten Umfang sicher als unkritisch erkannt. Die Auswertung der Gassensorsignale liefert eine Unterscheidung von Nitrat- und Peroxid-basierten Sprengstoffen sowie von nicht-explosiven Substanzen.
In the presented project, new approaches for the prevention of hand movements leading to hazards and for non-contact detection of fingers are intended to permit comprehensive and economical protection on circular saws. The basic principles may also be applied to other machines with manual loading and/or unloading. Two new detection principles are explained. The first is the distinction between skin and wood or other material by spectral analysis in the near infrared region. Using LED and photodiodes it is possible to detect fingers and hands reliable. With a kind of light curtain the intrusion into the dangerous zone near the blade can be prevented. The second principle is video image processing to detect persons, arms and fingers. In the first stage of development the detection of upper limb extremities within a defined hazard area by means of a computer based video image analysis is investigated.
Im Rahmen der Forschungsprojekte FeGeb und SPAI wurden bei zahlreichen Probanden Hautproben an mehreren Stellen des Gesichts, sowie der Arme und Hände, mit einem Nahinfrarot-Spektrometer (NIR, auch „Short Wave Infrared“, SWIR) erfasst und die Gesichter der Probanden zusätzlich mit einer hochwertigen Farb-Kamera, sowie einem selbst entwickelten multispektralen NIR-Kamerasystem aus mehreren Perspektiven aufgenommen. Vorrangiges Ziel dieser Messreihe war es, die Robustheit des an der Hochschule entwickelten Verfahrens zur berührungslosen Hauterkennung mittels multispektraler Nahinfrarotsensorik nachzuweisen. Haut ist im nahinfraroten Spektralbereich unabhängig von Geschlecht, Alter und Hauttyp sehr gut von anderen Materialien unterscheidbar. Weiterhin konnte mit Hilfe der so aufgenommenen Daten ein Klassifikator trainiert werden, der auch „Fälschungen“ wie beispielsweise Latexmasken zuverlässig von echter Haut unterscheiden kann.
Ein Teil der aus dieser Messreihe entstandenen Datenbank kann zum Download angefordert und für wissenschaftliche und akademische Zwecke in Forschung und Lehre kostenfrei verwendet werden.
A Fourier scatterometry setup is evaluated to recover the key parameters of optical phase gratings. Based on these parameters, systematic errors in the printing process of two-photon polymerization (TPP) gray-scale lithography three-dimensional printers can be compensated, namely tilt and curvature deviations. The proposed setup is significantly cheaper than a confocal microscope, which is usually used to determine calibration parameters for compensation of the TPP printing process. The grating parameters recovered this way are compared to those obtained with a confocal microscope. A clear correlation between confocal and scatterometric measurements is first shown for structures containing either tilt or curvature. The correlation is also shown for structures containing a mixture of tilt and curvature errors (squared Pearson coefficient r2 = 0.92). This compensation method is demonstrated on a TPP printer: a diffractive optical element printed with correction parameters obtained from Fourier scatterometry shows a significant reduction in noise as compared to the uncompensated system. This verifies the successful reduction of tilt and curvature errors. Further improvements of the method are proposed, which may enable the measurements to become more precise than confocal measurements in the future, since scatterometry is not affected by the diffraction limit.
Kollaborative Industrieroboter werden für produzierende Unternehmen immer kosteneffizienter. Während diese Systeme für den menschlichen Mitarbeiter eine große Hilfe sein können, stellen sie gleichzeitig ein ernstes Gesundheitsrisiko dar, wenn die zwingend notwendigen Sicherheitsmaßnahmen nur unzureichend umgesetzt werden. Herkömmliche Sicherheitseinrichtungen wie Zäune oder Lichtvorhänge bieten einen guten Schutz, aber solch statische Schutzvorrichtungen sind in neuen, hochdynamischen Arbeitsszenarien problematisch.
Im Forschungsprojekt BeyondSPAI wurde ein Funktionsmuster eines Multisensorsystems zur Absicherung solcher dynamischer Arbeitsszenarien entworfen, implementiert und im Feld getestet. Kern des Systems ist eine robuste optische Materialklassifikation, die mit Hilfe eines intelligenten InGaAs-Kamerasystems Haut von anderen typischen Werkstückoberflächen (z.B. Holz, Metalle od. Kunststoffe) unterscheiden kann. Diese einzigartige Eigenschaft wird genutzt, um menschliche Mitarbeiter zuverlässig zu erkennen, so dass ein konventioneller Roboter in Folge als personenbewusster Cobot arbeiten kann.
Das System ist modular und kann leicht mit weiteren Sensoren verschiedenster Art erweitert werden. Es kann an verschiedene Marken von Industrierobotern angepasst werden und lässt sich schnell an bestehenden Robotersystemen integrieren. Die vier vom System bereitgestellten Sicherheitsausgänge können dazu verwendet werden - abhängig von der durchdrungenen Überwachungszone - entweder eine Warnung auszugeben, die Bewegung des Roboters auf eine sichere Geschwindigkeit zu verlangsamen, oder den Roboter sicher anzuhalten. Sobald alle Zonen wieder als „eindeutig frei von Personen“ identifiziert sind, kann der Roboter wieder beschleunigen, seine ursprüngliche Bewegung wiederaufnehmen und die Arbeit fortsetzen.
Der Asiatische Laubholzbockkäfer (Anoplophora glabripennis, kurz: ALB) ist ein Bockkäfer, der 2001 seinen Weg nach Europa fand. Er ist als Quarantäneschaderreger eingestuft und muss in Europa bekämpft werden. Eine der Möglichkeiten zum Aufspüren befallener Bäume ist der Einsatz von Spürhunden. Die Einstufung des ALB als Quarantäneschädling bringt große Probleme bei der Verwendung von Trainingsmaterial mit sich. Da es sich zudem um biologisches Material handelt, das geruchchemisch Änderungen und Variationen unterworfen ist, und da die für den Hund relevanten Geruchsstoffe nicht bekannt sind, ist es häufig schwierig, geeignete und frische Geruchsträger als Trainingshilfsmittel zur Verfügung zu stellen.
Intimate swabs taken for examination in sexual assault cases typically yield mixtures of sperm and epithelial cell types. While powerful, differential extraction protocols to overcome such cell type mixtures by separate lysis of epithelial cells and spermatozoa can still prove ineffective, in particular if only few sperm cells are present or if swabs contain sperm from more than one individual leading to complex low level DNA mixtures. A means to avoid such mixtures consists in the analysis of single micromanipulated sperm cells. However, the quantity of DNA from single sperm cells is not sufficient for conventional STR analysis. Here, we describe a simple method for micromanipulating individual sperm cells from intimate swabs and show that whole genome amplification can generate sufficient amounts of DNA from single cells for subsequent DNA profiling. We recovered over 80% of alleles of haploid autosomal STR profiles from the majority of individual sperm cells. Furthermore, we demonstrate that in mixtures of sperm from two contributors, Y-STR and X-STR profiles of individual sperm cells can be used to sort the haploid autosomal profiles to develop the diploid consensus STR profiles of the individual donors. Finally, by analysing single sperm cells from mock sexual assault swabs with one or two sperm donors, we showed that our protocols enabled the identification of the unknown male contributors.
Discrimination of Stressed and Non-Stressed Food-Related Bacteria Using Raman-Microspectroscopy

As the identification of microorganisms becomes more significant in industry, so does the utilization of microspectroscopy and the development of effective chemometric models for data analysis and classification. Since only microorganisms cultivated under laboratory conditions can be identified, but they are exposed to a variety of stress factors, such as temperature differences, there is a demand for a method that can take these stress factors and the associated reactions of the bacteria into account. Therefore, bacterial stress reactions to lifetime conditions (regular treatment, 25 °C, HCl, 2-propanol, NaOH) and sampling conditions (cold sampling, desiccation, heat drying) were induced to explore the effects on Raman spectra in order to improve the chemometric models. As a result, in this study nine food-relevant bacteria were exposed to seven stress conditions in addition to routine cultivation as a control. Spectral alterations in lipids, polysaccharides, nucleic acids, and proteins were observed when compared to normal growth circumstances without stresses. Regardless of the involvement of several stress factors and storage times, a model for differentiating the analyzed microorganisms from genus down to strain level was developed. Classification of the independent training dataset at genus and species level for Escherichia coli and at strain level for the other food relevant microorganisms showed a classification rate of 97.6%.
Surface-enhanced Raman spectroscopy (SERS) with subsequent chemometric evaluation was performed for the rapid and non-destructive differentiation of seven important meat-associated microorganisms, namely Brochothrix thermosphacta DSM 20171, Pseudomonas fluorescens DSM 4358, Salmonella enterica subsp. enterica sv. Enteritidis DSM 14221, Listeria monocytogenes DSM 19094, Micrococcus luteus DSM 20030, Escherichia coli HB101 and Bacillus thuringiensis sv. israelensis DSM 5724. A simple method for collecting spectra from commercial paper-based SERS substrates without any laborious pre-treatments was used. In order to prepare the spectroscopic data for classification at genera level with a subsequent chemometric evaluation consisting of principal component analysis and discriminant analysis, a pre-processing method with spike correction and sum normalisation was performed. Because of the spike correction rather than exclusion, and therefore the use of a balanced data set, the multivariate analysis of the data is significantly resilient and meaningful. The analysis showed that the differentiation of meat-associated microorganisms and thereby the detection of important meat-related pathogenic bacteria was successful on genera level and a cross-validation as well as a classification of ungrouped data showed promising results, with 99.5 % and 97.5 %, respectively.
Hydrophilic surface-enhanced Raman spectroscopy (SERS) substrates were prepared by a combination of TiO2-coatings of aluminium plates through a direct titanium tetraisopropoxide (TTIP) coating and drop coated by synthesised gold nanoparticles (AuNPs). Differences between the wettability of the untreated substrates, the slowly dried Ti(OH)4 substrates and calcinated as well as plasma treated TiO2 substrates were analysed by water contact angle (WCA) measurements. The hydrophilic behaviour of the developed substrates helped to improve the distribution of the AuNPs, which reflects in overall higher lateral SERS enhancement. Surface enhancement of the substrates was tested with target molecule rhodamine 6G (R6G) and a fibre-coupled 638 nm Raman spectrometer. Additionally, the morphology of the substrates was characterised using scanning electron microscopy (SEM) and Raman microscopy. The studies showed a reduced influence of the coffee ring effect on the particle distribution, resulting in a more broadly distributed edge region, which increased the spatial reproducibility of the measured SERS signal in the surface-enhanced Raman mapping measurements on mm scale.
Design of an Active Multispectral SWIR Camera System for Skin Detection and Face Verification

Biometric face recognition is becoming more frequently used in different application scenarios. However, spoofing attacks with facial disguises are still a serious problem for state of the art face recognition algorithms. This work proposes an approach to face verification based on spectral signatures of material surfaces in the short wave infrared (SWIR) range. They allow distinguishing authentic human skin reliably from other materials, independent of the skin type. We present the design of an active SWIR imaging system that acquires four-band multispectral image stacks in real-time. The system uses pulsed small band illumination, which allows for fast image acquisition and high spectral resolution and renders it widely independent of ambient light. After extracting the spectral signatures from the acquired images, detected faces can be verified or rejected by classifying the material as "skin" or "no-skin". The approach is extensively evaluated with respect to both acquisition and classification performance. In addition, we present a database containing RGB and multispectral SWIR face images, as well as spectrometer measurements of a variety of subjects, which is used to evaluate our approach and will be made available to the research community by the time this work is published.
The human gut microbiota harbors untapped potential for biotechnological applications. Within the phylum of Bacteroidota, Phocaeicola vulgatus stands out as a promising candidate for sustainable production of key platform chemicals like succinate. However, genetic engineering of Phocaeicola sp. remains challenging due to its intricate molecular landscape. This study lays the groundwork for manipulating the central carbon pathways in Phocaeicola vulgatus, offering insights into overcoming genetic hurdles for increased succinate yields.
Due to their user-friendliness and reliability, biometric systems have taken a central role in everyday digital identity management for all kinds of private, financial and governmental applications with increasing security requirements. A central security aspect of unsupervised biometric authentication systems is the presentation attack detection (PAD) mechanism, which defines the robustness to fake or altered biometric features. Artifacts like photos, artificial fingers, face masks and fake iris contact lenses are a general security threat for all biometric modalities. The Biometric Evaluation Center of the Institute of Safety and Security Research (ISF) at the University of Applied Sciences Bonn-Rhein-Sieg has specialized in the development of a near-infrared (NIR)-based contact-less detection technology that can distinguish between human skin and most artifact materials. This technology is highly adaptable and has already been successfully integrated into fingerprint scanners, face recognition devices and hand vein scanners. In this work, we introduce a cutting-edge, miniaturized near-infrared presentation attack detection (NIR-PAD) device. It includes an innovative signal processing chain and an integrated distance measurement feature to boost both reliability and resilience. We detail the device’s modular configuration and conceptual decisions, highlighting its suitability as a versatile platform for sensor fusion and seamless integration into future biometric systems. This paper elucidates the technological foundations and conceptual framework of the NIR-PAD reference platform, alongside an exploration of its potential applications and prospective enhancements.
