Background: Human mesenchymal stem cells (hMSCs) have shown their multipotential including differentiating towards endothelial and smooth muscle cell lineages, which triggers a new interest for using hMSCs as a putative source for cardiovascular regenerative medicine. Our recent publication has shown for the first time that purinergic 2 receptors are key players during hMSC differentiation towards adipocytes and osteoblasts. Purinergic 2 receptors play an important role in cardiovascular function when they bind to extracellular nucleotides. In this study, the possible functional role of purinergic 2 receptors during MSC endothelial and smooth muscle differentiation was investigated. Methods and Results: Human MSCs were isolated from liposuction materials. Then, endothelial and smooth muscle-like cells were differentiated and characterized by specific markers via Reverse Transcriptase-PCR (RT-PCR), Western blot and immunochemical stainings. Interestingly, some purinergic 2 receptor subtypes were found to be differently regulated during these specific lineage commitments: P2Y4 and P2Y14 were involved in the early stage commitment while P2Y1 was the key player in controlling MSC differentiation towards either endothelial or smooth muscle cells. The administration of natural and artificial purinergic 2 receptor agonists and antagonists had a direct influence on these differentiations. Moreover, a feedback loop via exogenous extracellular nucleotides on these particular differentiations was shown by apyrase digest. Conclusions: Purinergic 2 receptors play a crucial role during the differentiation towards endothelial and smooth muscle cell lineages. Some highly selective and potent artificial purinergic 2 ligands can control hMSC differentiation, which might improve the use of adult stem cells in cardiovascular tissue engineering in the future.
Graph drawing with spring embedders employs a V x V computation phase over the graph's vertex set to compute repulsive forces. Here, the efficacy of forces diminishes with distance: a vertex can effectively only influence other vertices in a certain radius around its position. Therefore, the algorithm lends itself to an implementation using search data structures to reduce the runtime complexity. NVIDIA RT cores implement hierarchical tree traversal in hardware. We show how to map the problem of finding graph layouts with force-directed methods to a ray tracing problem that can subsequently be implemented with dedicated ray tracing hardware. With that, we observe speedups of 4x to 13x over a CUDA software implementation.
Extremophiles are optimal models in experimentally addressing questions about the effects of cosmic radiation on biological systems. The resistance to high charge energy (HZE) particles, and helium (He) ions and iron (Fe) ions (LET at 2.2 and 200 keV/µm, respectively, until 1000 Gy), of spores from two thermophiles, Bacillushorneckiae SBP3 and Bacilluslicheniformis T14, and two psychrotolerants, Bacillus sp. A34 and A43, was investigated. Spores survived He irradiation better, whereas they were more sensitive to Fe irradiation (until 500 Gy), with spores from thermophiles being more resistant to irradiations than psychrotolerants. The survived spores showed different germination kinetics, depending on the type/dose of irradiation and the germinant used. After exposure to He 1000 Gy, D-glucose increased the lag time of thermophilic spores and induced germination of psychrotolerants, whereas L-alanine and L-valine increased the germination efficiency, except alanine for A43. FTIR spectra showed important modifications to the structural components of spores after Fe irradiation at 250 Gy, which could explain the block in spore germination, whereas minor changes were observed after He radiation that could be related to the increased permeability of the inner membranes and alterations of receptor complex structures. Our results give new insights on HZE resistance of extremophiles that are useful in different contexts, including astrobiology.
This work thoroughly investigates a semi-Lagrangian lattice Boltzmann (SLLBM) solver for compressible flows. In contrast to other LBM for compressible flows, the vertices are organized in cells, and interpolation polynomials up to fourth order are used to attain the off-vertex distribution function values. Differing from the recently introduced Particles on Demand (PoD) method , the method operates in a static, non-moving reference frame. Yet the SLLBM in the present formulation grants supersonic flows and exhibits a high degree of Galilean invariance. The SLLBM solver allows for an independent time step size due to the integration along characteristics and for the use of unusual velocity sets, like the D2Q25, which is constructed by the roots of the fifth-order Hermite polynomial. The properties of the present model are shown in diverse example simulations of a two-dimensional Taylor-Green vortex, a Sod shock tube, a two-dimensional Riemann problem and a shock-vortex interaction. It is shown that the cell-based interpolation and the use of Gauss-Lobatto-Chebyshev support points allow for spatially high-order solutions and minimize the mass loss caused by the interpolation. Transformed grids in the shock-vortex interaction show the general applicability to non-uniform grids.
Turbulent compressible flows are traditionally simulated using explicit Eulerian time integration applied to the Navier-Stokes equations. However, the associated Courant-Friedrichs-Lewy condition severely restricts the maximum time step size. Exploiting the Lagrangian nature of the Boltzmann equation's material derivative, we now introduce a feasible three-dimensional semi-Lagrangian lattice Boltzmann method (SLLBM), which elegantly circumvents this restriction. Previous lattice Boltzmann methods for compressible flows were mostly restricted to two dimensions due to the enormous number of discrete velocities needed in three dimensions. In contrast, this Rapid Communication demonstrates how cubature rules enhance the SLLBM to yield a three-dimensional velocity set with only 45 discrete velocities. Based on simulations of a compressible Taylor-Green vortex we show that the new method accurately captures shocks or shocklets as well as turbulence in 3D without utilizing additional filtering or stabilizing techniques, even when the time step sizes are up to two orders of magnitude larger compared to simulations in the literature. Our new method therefore enables researchers for the first time to study compressible turbulent flows by a fully explicit scheme, whose range of admissible time step sizes is only dictated by physics, while being decoupled from the spatial discretization.
This paper introduces FaceHaptics, a novel haptic display based on a robot arm attached to a head-mounted virtual reality display. It provides localized, multi-directional and movable haptic cues in the form of wind, warmth, moving and single-point touch events and water spray to dedicated parts of the face not covered by the head-mounted display.The easily extensible system, however, can principally mount any type of compact haptic actuator or object. User study 1 showed that users appreciate the directional resolution of cues, and can judge wind direction well, especially when they move their head and wind direction is adjusted dynamically to compensate for head rotations. Study 2 showed that adding FaceHaptics cues to a VR walkthrough can significantly improve user experience, presence, and emotional responses.
Risk-based Authentication (RBA) is an adaptive security measure that improves the security of password-based authentication by protecting against credential stuffing, password guessing, or phishing attacks. RBA monitors extra features during login and requests for an additional authentication step if the observed feature values deviate from the usual ones in the login history. In state-of-the-art RBA re-authentication deployments, users receive an email with a numerical code in its body, which must be entered on the online service. Although this procedure has a major impact on RBA's time exposure and usability, these aspects were not studied so far.
We introduce two RBA re-authentication variants supplementing the de facto standard with a link-based and another code-based approach. Then, we present the results of a between-group study (N=592) to evaluate these three approaches. Our observations show with significant results that there is potential to speed up the RBA re-authentication process without reducing neither its security properties nor its security perception. The link-based re-authentication via "magic links", however, makes users significantly more anxious than the code-based approaches when perceived for the first time. Our evaluations underline the fact that RBA re-authentication is not a uniform procedure. We summarize our findings and provide recommendations.
Risk-based Authentication (RBA) is an adaptive security measure to strengthen password-based authentication. RBA monitors additional features during login, and when observed feature values differ significantly from previously seen ones, users have to provide additional authentication factors such as a verification code. RBA has the potential to offer more usable authentication, but the usability and the security perceptions of RBA are not studied well.
We present the results of a between-group lab study (n=65) to evaluate usability and security perceptions of two RBA variants, one 2FA variant, and password-only authentication. Our study shows with significant results that RBA is considered to be more usable than the studied 2FA variants, while it is perceived as more secure than password-only authentication in general and comparably secure to 2FA in a variety of application types. We also observed RBA usability problems and provide recommendations for mitigation. Our contribution provides a first deeper understanding of the users' perception of RBA and helps to improve RBA implementations for a broader user acceptance.
The development of sustainable, environmentally friendly insulation materials with a reduced carbon footprint is attracting increased interest. One alternative to conventional insulation materials are foamed geopolymers. Similar to foamed concrete, the mechanical properties of geopolymer foams can also be improved by using fibers for reinforcement. This paper presents an overview of the latest research findings in the field of fiber-reinforced geopolymer foam concrete with special focus on natural fibers reinforcement. Furthermore, some basic and background information of natural fibers and geopolymer foams are reported. In most of the research, foams are produced either through chemical foaming with hydrogen peroxide or aluminum powder, or through mechanical foaming which includes a foaming agent. However, previous reviews have not sufficiently addresses the fabrication of geopolymer foams by syntactic foams. Finally, recent efforts to reduce the fiber degradation in geopolymer concrete are discussed along with challenges for natural fiber reinforced-geopolymer foam concrete.
Human and robot tasks in household environments include actions such as carrying an object, cleaning a surface, etc. These tasks are performed by means of dexterous manipulation, and for humans, they are straightforward to accomplish. Moreover, humans perform these actions with reasonable accuracy and precision but with much less energy and stress on the actuators (muscles) than the robots do. The high agility in controlling their forces and motions is actually due to "laziness", i.e. humans exploit the existing natural forces and constraints to execute the tasks.
The above-mentioned properties of the human lazy strategy motivate us to relax the problem of controlling robot motions and forces, and solve it with the help of the environment. Therefore, in this work, we developed a lazy control strategy, i.e. task specification models and control architectures that relax several aspects of robot control by exploiting prior knowledge about the task and environment. The developed control strategy is realized in four different robotics use cases. In this work, the Popov-Vereshchagin hybrid dynamics solver is used as one of the building blocks in the proposed control architectures. An extension of the solver’s interface with the artificial Cartesian force and feed-forward joint torque task-drivers is proposed in this thesis.
To validate the proposed lazy control approach, an experimental evaluation was performed in a simulation environment and on a real robot platform.
Striated muscle contraction is regulated by the translocation of troponin-tropomyosin strands over the thin filament surface. Relaxation relies partly on highly-favorable, conformation-dependent electrostatic contacts between actin and tropomyosin, which position tropomyosin such that it impedes actomyosin associations. Impaired relaxation and hypercontractile properties are hallmarks of various muscle disorders. The α-cardiac actin M305L hypertrophic cardiomyopathy-causing mutation lies near residues that help confine tropomyosin to an inhibitory position along thin filaments. Here, we investigate M305L actin in vivo, in vitro, and in silico to resolve emergent pathological properties and disease mechanisms. Our data suggest the mutation reduces actin flexibility and distorts the actin-tropomyosin electrostatic energy landscape that, in muscle, result in aberrant contractile inhibition and excessive force. Thus, actin flexibility may be required to establish and maintain interfacial contacts with tropomyosin as well as facilitate its movement over distinct actin surface features and is, therefore, likely necessary for proper regulation of contraction.
Exploring Future Work - Co-Designing a Human-robot Collaboration Environment for Service Domains

There has been increasing interest in the application of humanoid robots in service domains like retail or care homes in recent years. Here, most use cases focus on serving customer needs autonomously. Frequently, human intervention becomes necessary to support the robot in exceptional situations. However, direct intervention of service operators is often not possible and requires specialized personnel. In a co-design process with 13 service operators from a pharmacy, we designed a remote working environment for human-robot collaboration that enables first-time experiences and collaboration with robots. Five participants took part in an assessment study and reported on their experiences about the utility, usability and user experience. Results show that participants were able to control and train the robot through the remote control environment. We discuss implications of our results for future work in service domains and emphasize a shift of focus from full robot automatization to human-robot collaboration forms.

Insights from an Exergame-Based Training System for People with Dementia and Their Caregivers

There has been increasing interest in designing for dementia in recent years. Empirical investigation is now needed of the long-term role of caregivers in appropriating ICTs into the complex daily life of people with dementia (PwD). We present here the outcomes of a 4-month evaluation of the individual, social and institutional impact of a videogame-based training system. The everyday behavior and interactions of 52 PwD and 25 caregivers was studied qualitatively, focusing on the role played by caregivers in integrating the system into daily routines. Our results indicate that the successful appropriation of ICT for PwD depends partly on the physical, cognitive and social benefits for PwD, but especially on the added value perceived by their social care-network. We discuss the need for design in dementia to develop more socially embedded innovations that can address the social actors involved and thus contribute to practical solutions for professional and private care.

The ongoing digitisation in everyday working life means that ever larger amounts of personal data of employees are processed by their employers. This development is particularly problematic with regard to employee data protection and the right to informational self-determination. We strive for the use of company Privacy Dashboards as a means to compensate for missing transparency and control. For conceptual design we use among other things the method of mental models. We present the methodology and first results of our research. We highlight the opportunities that such an approach offers for the user-centred development of Privacy Dashboards.
In forensic DNA profiling, the occurrence of complex mixed profiles is currently a common issue. Cases involving intimate swabs or skin flake tape liftings are prone to mixed profiles, because of more than one donor contributing to a DNA sample. By DNA profiling of single spermatozoa and skin flakes, problems associated with mixed profile could ideally be overcome. However, PCR is not a sensitive enough method to generate DNA profiles by STRs on single cells. Moreover, high quality intact DNA is required, but is not always available in skin flakes due to degradation. Additionally, single skin flakes are difficult to discriminate from other similar looking particles on tape liftings used to secure DNA samples from evidence. The main purpose of this study was to develop a method that enables DNA profiling of single sperm cells and skin flakes. After studying multiple whole genome amplification (WGA) protocols, REPLI-g Single Cell WGA was selected due to its suitability in the pre-amplification step of template DNA. Micromanipulation was used to isolate single spermatozoa. Furthermore, micromanipulation in combination with REPLI-g Single Cell WGA resulted in successful DNA profiling of single spermatozoa by using autosomal STRs as well as X- and Y-chromosomal STRs. The single spermatozoa DNA profiling method described in this thesis was successfully used to identify male contributors from mock intimate swabs with a mixture of semen from multiple male contributors. Different dyes were analysed to develop a staining method to discriminate skin flakes from other particles including particles such as those from hair cosmetic products. From all dyes tested, Orange G was the only dye which successfully discriminated skin flakes from hair product particles. Also, an alkaline based lysis protocol was developed that allowed PCR to be carried out directly on the lysates of single skin flakes. Furthermore, REPLI-g Single Cell WGA was tested on single skin flakes. In contrast to the single spermatozoa, REPLI-g Single Cell WGA was not successful in DNA profiling of single skin flakes. The single skin flake DNA profiling method described in this thesis was successfully used in correctly identifying contributors from mock mixed DNA evidence. Additionally, a small amplicon-based NGS method was tested on single skin flakes. Compared to the PCR and CE approach, the small amplicon-based NGS method improved DNA profiling of single skin flakes, giving a significant increase in allele recovery. In conclusion, this study shows circumventing mixtures is possible by DNA profiling of single spermatozoa, using micromanipulation and WGA. Furthermore, DNA profiling of single skin flakes has been improved by the staining of tape liftings methodology with Orange G, alkaline lysis, direct-PCR and a small amplicon-based NGS approach. Nonetheless, future work is required to assess the performance of the single spermatozoa method on mock swabs with more diluted semen. Also, commercially available NGS kits should be tested with single skin flakes and compared with the in-house NGS method.
Who do you trust: Peers or Technology? A conjoint analysis about computational reputation mechanisms

Peer-to-peer sharing platforms are taking over an increasingly important role in the platform economy due to their sustainable business model. By sharing private goods and services, the challenge arises to build trust between peers online mostly without any kind of physical presence. Peer rating has been proven as an important mechanism. In this paper, we explore the concept called Trust Score, a computational rating mechanism adopted from car telematics, which can play a similar role in carsharing. For this purpose, we conducted a conjoint analysis where 77 car owners chose between fictitious user profiles. Our results show that in our experiment the telemetric-based score slightly outperforms the peer rating in the decision process, while the participants perceived the peer rating more helpful in retrospect. Further, we discuss potential benefits with regard to existing shortcomings of user rating, but also various concerns that should be considered in concepts like telemetric-based reputation mechanism that supplements existing trust factors such as user ratings.
Background: While work-related rumination increases the risk of acute stressors developing into chronic load reactions and adverse health, mental detachment has been suggested as a way to interrupt this chain. Despite the importance of mentally detaching from work during leisure time, workers seem to struggle to disengage and, instead, experience the constant mental representation of work-related stressors, regardless of their absence. Those who struggle with work-related rumination could benefit from an easy-access intervention that fosters mental detachment by promoting recreational activities. Especially during vacations, workers appear to naturally engage in sufficient recovery activities; however, this beneficial behaviour is not sustained. The smartphone app-based intervention “Holidaily” promotes recovery behaviour and, thus, mental detachment from work with the intension of extending the beneficial effects of workers’ vacations into their daily working life.
Methods: This randomised-controlled trial (RCT) evaluates the efficacy of “Holidaily”. The Holidaily app is a German stand-alone program for mobile devices with either Android/iOS operating systems. The sample includes workers, who are awaiting to go on vacation and are randomly assigned to either the intervention (IG) or a waitlist-control group (CG). The IG receives two weeks pre-vacation access to Holidaily, while the CG receives access two weeks post-vacation. On a daily basis participants in the IG are provided with three options promoting recreational activities and beneficial recovery experiences. Online questionnaires are distributed to all participants at several timepoints. The primary outcome measure assesses participants’ work-related rumination (Irritation Scale). A significant difference two weeks post-vacation is expected, favouring the IG. Secondary outcomes include symptoms of depression, insomnia severity, emotional exhaustion, thinking about work, recovery experiences, vacation specifics, work and personal characteristics. To help explain the intervention’s effect, explorative analyses will investigate the mediation properties of the frequency of engaging in recreational activities and the moderation properties of Holidaily users’ experiences.
Discussion: If successful, workers will maintain their recovery behaviour beyond their vacation into daily working life. Findings could, therefore, provide evidence for low-intensity interventions that could be very valuable from a public-health perspective. App-based interventions have greater reach; hence, more workers might access preventative tools to protect themselves from developing adverse health effects linked to work-related rumination. Further studies will still be needed to investigate whether the vacation phenomenon of “lots of fun quickly gone” can be defied and long-term benefits attained.
This paper presents groupware to study group behavior while conducting a creative task on large, high-resolution displays. Moreover, we present the results of a between-subjects study. In the study, 12 groups with two participants each prototyped a 2D level on a 7m x 2.5m large, high-resolution display using tablet-PCs for interaction. Six groups underwent a condition where group members had equal roles and interaction possibilities. Another six groups worked in a condition where group members had different roles: level designer and 2D artist. The results revealed that in the different roles condition, the participants worked significantly more tightly and created more assets. We could also detect some shortcomings for that configuration. We discuss the gained insights regarding system configuration, groupware interfaces, and groups behavior.

This work presents the development of a measuring system for the quality control of ultrapure water. The new systems combines ozonation and UV radiation for the oxidation of organic substances. The change in conductivity caused by the oxidation is furthermore correlated with the TOC of the solution.
Evaluation of a Multi-Layer 2.5D display in comparison to conventional 3D stereoscopic glasses

In this paper we propose and evaluate a custom-build projection-based multilayer 2.5D display, consisting of three layers of images, and compare performance to a stereoscopic 3D display. Stereoscopic vision can increase the involvement and enhance game experience, however may induce possible side effects, e.g. motion sickness and simulator sickness. To overcome the disadvantage of multiple discrete depths, in our system perspective rendering and head-tracking is used. A study was performed to evaluate this display with 20 participants playing custom-designed games. The results indicated that the multi-layer display caused fewer side effects than the stereoscopic display and provided good usability. The participants also stated a better or equal spatial perception, while the cognitive load stayed the same.
Any political phenomenon can only be properly understood in its broader con-text. Questions of international cooperation are thus necessarily framed by his-torical processes and relations of power. We therefore start our first discussion with an examination of the global ‘status quo’ and embed the topic of this pub-lication, ODA graduation, into the shifting world order, analysing current roles and settings in international relations and identifying changes in positions, sta-tus and categories. What are the overarching issues determining world politics and who are the old and the new actors driving them? What is the impact of these global shifts on international cooperation, especially development coop-eration? Of what relevance are roles, status and categories and what is the im-pact of changes in positions and relations? What challenges face multilateralism and what ways exist to maintain and renew strategic partnerships and shared values?
Mendelian diseases of dysregulated canonical NF-κB signaling: From immunodeficiency to inflammation

The ongoing coronavirus disease 2019 (COVID-19) pandemic threatens global health thereby causing unprecedented social, economic, and political disruptions. One way to prevent such a pandemic is through interventions at the human-animal-environment interface by using an integrated One Health (OH) approach. This systematic literature review documented the three coronavirus outbreaks, i.e. SARS, MERS, COVID-19, to evaluate the evolution of the OH approach, including the identification of key OH actions taken for prevention, response, and control.
The OH understandings identified were categorized into three distinct patterns: institutional coordination and collaboration, OH in action/implementation, and extended OH (i.e. a clear involvement of the environmental domain). Across all studies, OH was most often framed as OH in action/implementation and least often in its extended meaning. Utilizing OH as institutional coordination and collaboration and the extended OH both increased over time. OH actions were classified into twelve sub-groups and further categorized as classical OH actions (i.e. at the human-animal interface), classical OH actions with outcomes to the environment, and extended OH actions.
The majority of studies focused on human-animal interaction, giving less attention to the natural and built environment. Different understandings of the OH approach in practice and several practical limitations might hinder current efforts to achieve the operationalization of OH by combining institutional coordination and collaboration with specific OH actions. The actions identified here are a valuable starting point for evaluating the stage of OH development in different settings. This study showed that by moving beyond the classical OH approach and its actions towards a more extended understanding, OH can unfold its entire capacity thereby improving preparedness and mitigating the impacts of the next outbreak.
Modern Monte-Carlo-based rendering systems still suffer from the computational complexity involved in the generation of noise-free images, making it challenging to synthesize interactive previews. We present a framework suited for rendering such previews of static scenes using a caching technique that builds upon a linkless octree. Our approach allows for memory-efficient storage and constant-time lookup to cache diffuse illumination at multiple hitpoints along the traced paths. Non-diffuse surfaces are dealt with in a hybrid way in order to reconstruct view-dependent illumination while maintaining interactive frame rates. By evaluating the visual fidelity against ground truth sequences and by benchmarking, we show that our approach compares well to low-noise path-traced results, but with a greatly reduced computational complexity, allowing for interactive frame rates. This way, our caching technique provides a useful tool for global illumination previews and multi-view rendering.

Apple replant disease (ARD) is a soil-borne disease, which is of particular importance for fruit tree nurseries and fruit growers. The disease manifests by a poor vegetative development, stunted growth, and reduced yield in terms of quantity and quality, if apple plants (usually rootstocks) are replanted several times at the same site. Genotype-specific differences in the reaction of apple plants to ARD are documented, but less is known about the genetic mechanisms behind this symptomatology. Recent transcriptome analyses resulted in a number of candidate genes possibly involved in the plant response. In the present study, the expression of 108 selected candidate genes was investigated in root and leaf tissue of four different apple genotypes grown in untreated ARD soil and ARD soil disinfected by γ-irradiation originating from two different sites in Germany. Thirty-nine out of the 108 candidate genes were differentially expressed in roots by taking a p-value of < 0.05 and a fold change of > 1.5 as cutoff. Sixteen genes were more than 4.5-fold upregulated in roots of plants grown in ARD soil. The four genes MNL2 (putative mannosidase); ALF5 (multi antimicrobial extrusion protein); UGT73B4 (uridine diphosphate (UDP)-glycosyltransferase 73B4), and ECHI (chitin-binding) were significantly upregulated in roots. These genes seem to be related to the host plant response to ARD, although they have never been described in this context before. Six of the highly upregulated genes belong to the phytoalexin biosynthesis pathway. Their genotype-specific gene expression pattern was consistent with the phytoalexin content measured in roots. The biphenyl synthase (BIS) genes were found to be useful as early biomarkers for ARD, because their expression pattern correlated well with the phenotypic reaction of the Malus genotypes investigated.
Evaluation and Optimization of IEEE802.11 multi-hop Backhaul Networks with Directional Antennas

A major problem for rural areas is the inaccessibility to affordable broadband Internet connections. In these areas distances are large, and digging a cable into the ground is extremely expensive, considering the small number of potential customers at the end of that cable. This leads to a digital divide, where urban areas enjoy a high-quality service at low cost, while rural areas suffer from the reverse.
This work is dedicated to an alternative technical approach aiming to reduce the cost for Internet Service Provider in rural areas: WiFi-based Long Distance networks. A set of significant contributions of technology related aspects of WiFi-based Long Distance networks is described in three different fields: Propagation on long distance Wi-Fi links, MAC-layer scheduling and Interference modeling and Channel Assignment with directional antennas.
For each field, the author composes and discusses the state-of-the-art. Afterwards, the author derives research questions and tackles several open issues to develop these kinds of networks further towards a suitable technology for the backhaul segment.
Optical gas sensors based on chiral-nematic liquid crystals (N* LCs) forming one-dimensional photonic crystals do not require electrical energy and have a considerable potential to supplement established types of sensors. A chiral-nematic phase with tunable selective reflection is induced in a nematic host LC by adding reactive chiral dopants. The selective chemical reaction between dopant and analyte is capable to vary the pitch length (the lattice constant) of the soft, self-assembled, one-dimensional photonic crystal. The progress of the ongoing chemical reaction can be observed even by naked eye because the color of the samples varies. In this work, we encapsulate the responsive N* LC in microscale polyvinylpyrrolidone (PVP) fibers via coaxial electrospinning. The sensor is, thus, given a solid form and has an improved stability against nonavoidable environmental influences. The reaction behavior of encapsulated and nonencapsulated N* LC toward a gaseous analyte is compared, systematically. Making use of the encapsulation is an important step to improve the applicability.
The ability to finely segment different instances of various objects in an environment forms a critical tool in the perception tool-box of any autonomous agent. Traditionally instance segmentation is treated as a multi-label pixel-wise classification problem. This formulation has resulted in networks that are capable of producing high-quality instance masks but are extremely slow for real-world usage, especially on platforms with limited computational capabilities. This thesis investigates an alternate regression-based formulation of instance segmentation to achieve a good trade-off between mask precision and run-time. Particularly the instance masks are parameterized and a CNN is trained to regress to these parameters, analogous to bounding box regression performed by an object detection network.
In this investigation, the instance segmentation masks in the Cityscape dataset are approximated using irregular octagons and an existing object detector network (i.e., SqueezeDet) is modified to regresses to the parameters of these octagonal approximations. The resulting network is referred to as SqueezeDetOcta. At the image boundaries, object instances are only partially visible. Due to the convolutional nature of most object detection networks, special handling of the boundary adhering object instances is warranted. However, the current object detection techniques seem to be unaffected by this and handle all the object instances alike. To this end, this work proposes selectively learning only partial, untainted parameters of the bounding box approximation of the boundary adhering object instances. Anchor-based object detection networks like SqueezeDet and YOLOv2 have a discrepancy between the ground-truth encoding/decoding scheme and the coordinate space used for clustering, to generate the prior anchor shapes. To resolve this disagreement, this work proposes clustering in a space defined by two coordinate axes representing the natural log transformations of the width and height of the ground-truth bounding boxes.
When both SqueezeDet and SqueezeDetOcta were trained from scratch, SqueezeDetOcta lagged behind the SqueezeDet network by a massive ≈ 6.19 mAP. Further analysis revealed that the sparsity of the annotated data was the reason for this lackluster performance of the SqueezeDetOcta network. To mitigate this issue transfer-learning was used to fine-tune the SqueezeDetOcta network starting from the trained weights of the SqueezeDet network. When all the layers of the SqueezeDetOcta were fine-tuned, it outperformed the SqueezeDet network paired with logarithmically extracted anchors by ≈ 0.77 mAP. In addition to this, the forward pass latencies of both SqueezeDet and SqueezeDetOcta are close to ≈ 19ms. Boundary adhesion considerations, during training, resulted in an improvement of ≈ 2.62 mAP of the baseline SqueezeDet network. A SqueezeDet network paired with logarithmically extracted anchors improved the performance of the baseline SqueezeDet network by ≈ 1.85 mAP.
In summary, this work demonstrates that if given sufficient fine instance annotated data, an existing object detection network can be modified to predict much finer approximations (i.e., irregular octagons) of the instance annotations, whilst having the same forward pass latency as that of the bounding box predicting network. The results justify the merits of logarithmically extracted anchors to boost the performance of any anchor-based object detection network. The results also showed that the special handling of image boundary adhering object instances produces more performant object detectors.
This study advances the research and methodological approach to measuring and understanding national-level destination competitiveness, sustainability and governance, by creating a model that could be of use for both developing and developed destinations. The study gives a detailed overview of the research field of measuring destination competitiveness and sustainability. It also identifies major predictors of destination competitiveness and sustainability and thereby presents destination researchers and practitioners with a useful list of priority areas, both from a global perspective and from the perspective of other similar destinations. Finally, the study identifies two major types of destination governance with implications for research, policy and practice across the destination life-cycle. The research deals with the analysis of the secondary data from the World Economic Forum Travel and Tourism Index (WEF T&T). Major types of destination governance and predictors of belonging to either one of the types, as well as inside cluster predictors have been extracted through a two-step cluster analysis. The results support the notion that a meaningful model of national-level destination governance needs to take into account different development levels of different destinations. The main limitation of the study is its typology creation approach, as it inevitably leads to simplifications.
Compliant manipulation is a crucial skill for robots when they are supposed to act as helping hands in everyday household tasks. Still, nowadays, those skills are hand-crafted by experts which frequently requires labor-intensive, manual parameter tuning. Moreover, some tasks are too complex to be specified fully using a task specification. Learning these skills, by contrast, requires a high number of costly and potentially unsafe interactions with the environment. We present a compliant manipulation approach using reinforcement learning guided by the Task Frame Formalism, a task specification method. This allows us to specify the easy to model knowledge about a task while the robot learns the unmodeled components by reinforcement learning. We evaluate the approach by performing a compliant manipulation task with a KUKA LWR 4+ manipulator. The robot was able to learn force control policies directly on the robot without using any simulation.
One of the primary current astrobiological goals is to understand the limits of microbial resistance to extraterrestrial conditions. Much attention is paid to ionizing radiation, since it can prevent the preservation and spread of life outside the Earth. The aim of this research was to study the impact of accelerated He ions (150 MeV/n, up to 1 kGy) as a component of the galactic cosmic rays on the black fungus C. antarcticus when mixed with Antarctic sandstones—the substratum of its natural habitat—and two Martian regolith simulants, which mimics two different evolutionary stages of Mars. The high dose of 1 kGy was used to assess the effect of dose accumulation in dormant cells within minerals, under long-term irradiation estimated on a geological time scale. The data obtained suggests that viable Earth-like microorganisms can be preserved in the dormant state in the near-surface scenario for approximately 322,000 and 110,000 Earth years within Martian regolith that mimic early and present Mars environmental conditions, respectively. In addition, the results of the study indicate the possibility of maintaining traces within regolith, as demonstrated by the identification of melanin pigments through UltraViolet-visible (UV-vis) spectrophotometric approach.
With the digital transformation, software systems have become an integral part of our society and economy. In every part of our life, software systems are increasingly utilized to, e.g., simplify housework or to optimize business processes. All these applications are connected to the Internet, which already includes millions of software services consumed by billions of people. Applications which process such a magnitude of users and data traffic requires to be highly scalable and are therefore denoted as Ultra Large Scale (ULS) systems. Roy Fielding has defined one of the first approaches which allows designing modern ULS software systems. In his doctoral thesis, Fielding introduced the architectural style Representational State Transfer (REST) which builds the theoretical foundation of the web. At present, the web is considered as the world's largest ULS system. Due to a large number of users and the significance of software for society and the economy, the security of ULS systems is another crucial quality factor besides high scalability.
Long-term variability of solar irradiance and its implications for photovoltaic power in West Africa

This paper addresses long-term changes in solar irradiance for West Africa (3° N to 20° N and 20° W to 16° E) and its implications for photovoltaic power systems. Here we use satellite irradiance (Surface Solar Radiation Data Set-Heliosat, Edition 2.1, SARAH-2.1) to derive photovoltaic yields. Based on 35 years of data (1983–2017) the temporal and regional variability as well as long-term trends of global and direct horizontal irradiance are analyzed. Furthermore, at four locations a detailed time series analysis is undertaken. The dry and the wet season are considered separately.
This paper addresses long-term historical changes in solar irradiance in West Africa (3 to 20° N and 20° W to 16° E) and the implications for photovoltaic systems. Based on 35 years of data (1983–2017), the temporal and regional variability as well as long-term trends in global and direct horizontal irradiance are analyzed. Furthermore, a detailed time series analysis is undertaken at four locations. According to the high spatial resolution SARAH-2.1 data record (0.05°×0.05°), solar irradiance is largest (up to a 300 W m−2 daily average) in the Sahara and the Sahel zone with a positive trend (up to 5 W m−2 per decade) and a lower temporal variability (<75 W m−2 between 1983 and 2017 for daily averages). In contrast, the solar irradiance is lower in southern West Africa (between 200 W m−2 and 250 W m−2) with a negative trend (up to −5 W m−2 per decade) and a higher temporal variability (up to 150 W m−2). The positive trend in the north is mostly connected to the dry season, whereas the negative trend in the south occurs during the wet season. Both trends show 95 % significance. Photovoltaic (PV) yields show a strong meridional gradient with the lowest values of around 4 kWh kWp−1 in southern West Africa and values of more than 5.5 kWh kWp−1 in the Sahara and Sahel zone.
Long-term variability of solar irradiance and its implications for photovoltaic power in West Africa

West Africa is one of the least developed regions in the world regarding the energy availability and energy security. Located close to the equator West Africa receives high amounts of global horizontal irradiance (GHI). Thus, solar power and especially photovoltaic (PV) systems seem to be a promising solution to provide electricity with low environmental impact. To plan and to dimension a PV power system climatological data for global horizontal irradiance (GHI) and its variability need to be taken into account. However, ground based measurements of irradiances are not available continuously and cover only a few discrete locations.
For a sustainable development the electricity sector needs to be decarbonized. In 2017 only 54% of the West African households had access to the electrical grid. Thus, renewable sources should play a major role for the development of the power sector in West Africa. Above all, solar power shows highest potential of renewable energy sources. However, it is highly variable, depending on the atmospheric conditions. This study addresses the challenges for a solar based power system in West Africa by analyzing the atmospheric variability of solar power. For this purpose, two aspects are investigated. In the first part, the daily power reduction due to atmospheric aerosols is quantified for different solar power technologies. Meteorological data at six ground-based stations is used to model photovoltaic and parabolic trough power during all mostly clear-sky days in 2006. A radiative transfer model is combined with solar power model. The results show, that the reduction due to aerosols can be up to 79% for photovoltaic and up to 100% for parabolic trough power plants during a major dust outbreak. Frequent dust outbreaks occurring in West Africa would cause frequent blackouts if sufficient storage capacities are not available. On average, aerosols reduce the daily power yields by 13% to 22% for photovoltaic and by 22% to 37% for parabolic troughs. For the second part, long-term atmospheric variability and trends of solar irradiance are analyzed and their impact on photovoltaic yields is examined for West Africa. Based on a 35-year satellite data record (1983 - 2017) the temporal and spatial variability and general trend are depicted for global and direct horizontal irradiances. Furthermore, photovoltaic yields are calculated on a daily basis. They show a strong meridional gradient with highest values of 5 kWh/kWp in the Sahara and Sahel zone and lowest values in southern West Africa (around 4 kWh/kWp). Thereby, the temporal variability is highest in southern West Africa (up to around 18%) and lowest in the Sahara (around 4.5%). This implies the need of a North-South grid development, to feed the increasing demand on the highly populated coast by solar power from the northern parts of West Africa. Additionally, global irradiances show a long-term positive trend (up to +5 W/m²/decade) in the Sahara and a negative trend (up to -5 W/m²/decade) in southern West Africa. If this trend is continuing, the spatial differences in solar power potential will increase in the future. This thesis provides a better understanding of the impact of atmospheric variability on solar power in a challenging environment like West Africa, characterized by the strong influence of the African monsoon. Thereby, the importance of aerosols is pointed out. Furthermore, long-term changes of irradiance are characterized concerning their implications for photovoltaic power.
Grasp verification is advantageous for autonomous manipulation robots as they provide the feedback required for higher level planning components about successful task completion. However, a major obstacle in doing grasp verification is sensor selection. In this paper, we propose a vision based grasp verification system using machine vision cameras, with the verification problem formulated as an image classification task. Machine vision cameras consist of a camera and a processing unit capable of on-board deep learning inference. The inference in these low-power hardware are done near the data source, reducing the robot's dependence on a centralized server, leading to reduced latency, and improved reliability. Machine vision cameras provide the deep learning inference capabilities using different neural accelerators. Although, it is not clear from the documentation of these cameras what is the effect of these neural accelerators on performance metrics such as latency and throughput. To systematically benchmark these machine vision cameras, we propose a parameterized model generator that generates end to end models of Convolutional Neural Networks(CNN). Using these generated models we benchmark latency and throughput of two machine vision cameras, JeVois A33 and Sipeed Maix Bit. Our experiments demonstrate that the selected machine vision camera and the deep learning models can robustly verify grasp with 97% per frame accuracy.
Cytokine-induced killer (CIK) cells are heterogeneous, major histocompatibility complex (MHC)-unrestricted T lymphocytes that have acquired the expression of several natural killer (NK) cell surface markers following the addition of interferon gamma (IFN-γ), OKT3 and interleukin-2 (IL-2). Treatment with CIK cells demonstrates a practical approach in cancer immunotherapy with limited, if any, graft versus host disease (GvHD) toxicity. CIK cells have been proposed and tested in many clinical trials in cancer patients by autologous, allogeneic or haploidentical administration. The possibility of combining them with specific monoclonal antibodies nivolumab and ipilimumab will further expand the possibility of their clinical utilization. Initially, phenotypic analysis was performed to explore CD3, CD4, CD56, PD-1 and CTLA-4 expression on CIK cells and PD-L1/PD-L2 expression on tumor cells. We further treated CIK cells with nivolumab and ipilimumab and measured the cytotoxicity of CIK cells cocultured to renal carcinoma cell lines, A-498 and Caki-2. We observed a significant decrease in viability of renal cell lines after treating with CIK cells (p < 0.0001) in comparison to untreated renal cell lines and anti-PD-1 or anti-CTLA-4 treatment had no remarkable effect on the viability of tumor cells. Using CCK-8, Precision Count Beads™ and Cell Trace™ violet proliferation assays, we proved significant increased proliferation of CIK cells in the presence of a combination of anti-PD-1 and anti-CTLA-4 antibodies compared to untreated CIK cells. The IFN-γ secretion increased significantly in the presence of A-498 and combinatorial blockade of PD-1 and CTLA-4 compared to nivolumab or ipilimumab monotreatment (p < 0.001). In conclusion, a combination of immune checkpoint inhibition with CIK cells augments cytotoxicity of CIK cells against renal cancer cells.
Object detectors have improved considerably in the last years by using advanced CNN architectures. However, many detector hyper-parameters are generally manually tuned, or they are used with values set by the detector authors. Automatic Hyper-parameter optimization has not been explored in improving CNN-based object detectors hyper-parameters. In this work, we propose the use of Black-box optimization methods to tune the prior/default box scales in Faster R-CNN and SSD, using Bayesian Optimization, SMAC, and CMA-ES. We show that by tuning the input image size and prior box anchor scale on Faster R-CNN mAP increases by 2% on PASCAL VOC 2007, and by 3% with SSD. On the COCO dataset with SSD there are mAP improvement in the medium and large objects, but mAP decreases by 1% in small objects. We also perform a regression analysis to find the significant hyper-parameters to tune.
Object detectors have improved considerably in the last years by using advanced Convolutional Neural Networks (CNNs) architectures. However, many detector hyper-parameters are not generally tuned, and they are used with values set by the detector authors. Blackbox optimization methods have gained more attention in recent years because of its ability to optimize the hyper-parameters of various machine learning algorithms and deep learning models. However, these methods are not explored in improving CNN-based object detector's hyper-parameters. In this research work, we propose the use of blackbox optimization methods such as Gaussian Process based Bayesian Optimization (BOGP), Sequential Model-based Algorithm Configuration (SMAC), and Covariance Matrix Adaptation Evolution Strategy (CMA-ES) to tune the hyper-parameters in Faster R-CNN and Single Shot MultiBox Detector (SSD). In Faster R-CNN, tuning the input image size, prior box anchor scales and ratios using BOGP, SMAC, and CMA-ES has increased the performance around 1.5% in terms of Mean Average Precision (mAP) on PASCAL VOC. Tuning the anchor scales of SSD has increased the mAP by 3% on PASCAL VOC and marine debris datasets. On the COCO dataset with SSD, mAP improvement is observed in the medium and large objects, but mAP decreases by 1% in small objects. The experimental results show that the blackbox optimization methods have proved to increase the mAP performance by optimizing the object detectors. Moreover, it has achieved better results than the hand-tuned configurations in most of the cases.
Reinforcement learning (RL) algorithms should learn as much as possible about the environment but not the properties of the physics engines that generate the environment. There are multiple algorithms that solve the task in a physics engine based environment but there is no work done so far to understand if the RL algorithms can generalize across physics engines. In this work, we compare the generalization performance of various deep reinforcement learning algorithms on a variety of control tasks. Our results show that MuJoCo is the best engine to transfer the learning to other engines. On the other hand, none of the algorithms generalize when trained on PyBullet. We also found out that various algorithms have a promising generalizability if the effect of random seeds can be minimized on their performance.
When a robotic agent experiences a failure while acting in the world, it should be possible to discover why that failure has occurred, namely to diagnose the failure. In this paper, we argue that the diagnosability of robot actions, at least in a classical sense, is a feature that cannot be taken for granted since it strongly depends on the underlying action representation. We specifically define criteria that determine the diagnosability of robot actions. The diagnosability question is then analysed in the context of a handle manipulation action, such that we discuss two different representations of the action – a composite policy with a learned success model for the action parameters, and a neural network-based monolithic policy – both of which exist on different sides of the diagnosability spectrum. Through this comparison, we conclude that composite actions are more suited to explicit diagnosis, but representations with less prior knowledge are more flexible. This suggests that model learning may provide balance between flexibility and diagnosability; however, data-driven diagnosis methods also need to be enhanced in order to deal with the complexity of modern robots.

After unzipping the archives, each database can be restored with the command

Autonomous driving enables new mobility concepts such as shared-autonomous services. Although significant re-search has been done on passenger-car interaction, work on passenger interaction with robo-taxis is still rare. In this paper, we tackle the question of how passengers experience robo-taxis as a service in real-life settings to inform the interaction design. We conducted a Wizard of Oz study with an electric vehicle where the driver was hidden from the passenger to simulate the service experience of a robo-taxi. 10 participants had the opportunity to use the simulated shared-autonomous service in real-life situations for one week. By the week's end, 33 rides were completed and recorded on video. Also, we flanked the study conducting interviews before and after with all participants. The findings provided insights into four design themes that could inform the service design of robo-taxis along the different stages including hailing, pick-up, travel, and drop-off.

Facial emotion recognition is the task to classify human emotions in face images. It is a difficult task due to high aleatoric uncertainty and visual ambiguity. A large part of the literature aims to show progress by increasing accuracy on this task, but this ignores the inherent uncertainty and ambiguity in the task. In this paper we show that Bayesian Neural Networks, as approximated using MC-Dropout, MC-DropConnect, or an Ensemble, are able to model the aleatoric uncertainty in facial emotion recognition, and produce output probabilities that are closer to what a human expects. We also show that calibration metrics show strange behaviors for this task, due to the multiple classes that can be considered correct, which motivates future work. We believe our work will motivate other researchers to move away from Classical and into Bayesian Neural Networks.
